| ObjectiveTo investigate the effects of docetaxel on the growth and apoptosis of tongue cancer cell line Tca8113.Materials and Methods1. The tongue Tca8113 cell line of human sequamous cell carcinoma was bought from Shanghai Institute of Cell Biology, Chinese Academy of Sciences.2.Docetaxel was bought from Franch Aventis Company.3.MTT method:Incubated Tca8113 cells were treated with docetaxel at different concentration(0.001,0.01,0.1,1,10μmol/L)and continued incubating for 12h,24h,48h respectively.The inhibitory rate of cell proliferation was evaluated by MTT, At the same time, morphology of cells were observed with inverted microscope and fluorescence microscope.4.Distribution of cell cycle and apoptosis were detected by flow cytometer(FCM).5.The data were analyzed by SPSS13.0 softeware. Measuring data were shown by the formula "x±s", Method is repeated measurement and analysis of variance.Statistically significant level was considered as "alpha equals 0.05".Results1.Growth inhibitory effect of docetaxel on Tca8113 cells:Following treatment with docetaxel at different concentration(0.001,0.01,0.1,1,10μmol/L), the proliferation of Tca8113 cells was inhibited significantly(P<0.01).The inhibitory rate of cells proliferation at the same conceration of docetaxel has statistical significance in different time(12h,24h,48h)(P<0.01).The results demonstated that docetaxel could inhibit the Tca8113 cells growth by time-and dose-dependment manners.2.Cell cycle detected by flow cytometer:the effects which Docetaxel at 0.1μmol/L concentration had on the tongue cells indicate that Docetaxel stagnated the cells during the G2/M period and with the extension of time, the proportion of the cells in G2/M period gradually increased. After 48 hours, the proportion of the cells in G2/M period accounted to (41.67±5.84)%,which showed a statistical difference comparing to the matched group and other two groups.(P< 0.01).3. Cell apoptosis rate tested by flow cytometry:the apoptosis rate of the tongue cancer cells tended to increase after Docetaxel at 0.1μmol/L concentration excerted effect on them separately for 12 hours,24 hours and 48 hours. The results showed a significant difference compared with the matched group (P< 0.01).Conclusions1.Docetaxel can inhibit the proliferation of Tca8113 cells in a time-and dose-dependment manners·2.Docetaxel can change distribution of Tca8113 cells cycle.lt has evident effect that makes cells arrest at G2/M phase.lt can induce apoptosis of Tca8113 cells. |
