Objective:To analysis the correlations among IGF-1,IL-17 in maternal & cord blood serum,IGF-1-protein in placenta, insulin resistance of gestational diabetes mellitus and fetal birth weight, and to explore the relationships of IGF-1,IL-17 on IR and fetal growth development.Methods:Select 30 cases with GDM and 12 normal pregnant women as the control group, both of them were cesarean full-term deliveries. We collected maternal fasting blood before the onset of delivery and cord venous blood immediately after birth. Assay IGF-1 and IL-17 in the serum by ELISA. We analyses the correlation among IGF-1, IL-17, HOMA-IR and fetal birth weight. Furthermore, the relationship between IGF-1 and IL-17 is discussed.We collected placental villous tissue on maternal side, to observe the pathological change in placental tissue by HE. We determine the expression of IGF-1-protein in placenta by immunohistochemisty and analysis its relationship with IGF-1 in maternal& cord serum, fetal birth weight and so on.Results:1.The IGF-1 levels in maternal serum of two groups were significant difference(P<0.05), and they were significantly different in the umbilical cord serum groups(P<0.01).2.The IL-17 content in maternal serum of the two pregnant women groups was significant difference (P<0.05), but in the two groups of cord serum showed ho significant difference(P>0.05).3.HOMA-IR and IGF-1 which in maternal serum was negatively correlated, but HOMA-IR was positively related with IL-17 in maternal serum, the IGF-1 and IL-17 of the maternal serum was negatively correlated.4.The birth weight between two groups were significantly different, the GDM group was significantly higher. Fetal birth weight and the IGF-1 of umbilical cord serum was positively correlated.5.The morphology in the two groups of placental tissue was different, in the GDM group placenta stem villi small arteries thickening and luminal stenosis, villous stromal vascular changes such as over-filling. The IGF-1-protein was expressed in the syncytiotrophoblast of the two groups, but the GDM group was higher, the difference was significant (P<0.01).The IGF-1 of placental expression was positively correlated with the cord serum one and birth weight.Conclusions:1.The IGF-1 in maternal and cord serum are two independent systems, the one in the maternal was correlated with IR and GDM. The IGF-1 was expressed in the placenta, suggesting that may be the main source of IGF-1 which in cord serum. The IGF-1 of cord serum is an important factor in the fetal growth and development.2. There are pathological changes in the placenta of GDM group, and affecting intrauterine fetal growth and development.3. The level of IGF-1 and IL-17 in maternal serum has some relevance, and they are involved in the occurrence of IR and GDM.
