Background and Objective:Systemic Inflammatory Response Syndrome (SIRS) is a group of clinical syndromes occurs when body loses control of inflammatory reaction under infection or non-infection because mediators of inflammation increase greatly after over activation of inflammatory cell. SIRS is the cause of Multiple Organ Dysfunction Syndrome (MODS).Acute pancreatitis is one of the important disease causing SIRS.CD14 is the special differentiation antigen on mononuclear macrophage surface.It is synthesized and secreted by mononuclear macrophage,and express many kinds of cytokines through mCD14 signaling pathway.Stimuli to inflamematory cell without mCD14 on LPS mainly mediated by sCD14. sCD14 is functional receptor which mediates endotoxin reaction.sCD14 can combine with LPS in cycling and elevate cell sensitivity to endotoxin, then from malignant cycling, finally multiorgan failure.Effect of active Emodin in Da Cheng Qi Tang is good in SIRS treatment.Emodin can protect intestinal mucosa,eliminate oxygen free radicle and reduce lung injure.At the same time emodin can lower endotoxin in rat blood of SIRS, reduce content of TNF in blood and intestinal tissue. In this study, the aim is to study the function of applied Da Cheng Qi Tang to SIRS in severe acute pancreatitis in SAP/SIRS rat peritoneal macrophage sCD14 through making the SAP/SIRS rat model. To investigate the change and possible mechanism of peritoneal macrophages of SIRS in severe acute pancreatitis. And provide a theoretical basis for clinical drug therapy.Methods:Build severe acute pancreatitis SIRS rat model.Including five droups:control group, SIRS group,traditional medicine group,western medicine group and integrated traditional and western medicine group.Take peritoneal lavage fluid. Centrifugalization in vitro and store in-80℃refrig 24h after modeling.Detect concentration of sCD14 by ELISA in different groups.Results:PMΦin different groups:integrated traditional and western medicine group is obviously lower than SIRS group(31.83±29.51 vs 272.82±40.68, P<0.05); integrated traditional and western medicine group is obviously lower than traditional medicine group and western medicine group respectively (31.83±29.51 vs 88.97±54.80, P<0.05; 31.83±29.51 vs 83.45±35.21, P<0.05);there is no difference between integrated traditional and western medicine group and control group (31.83±29.51 vs 31.30±24.30, P>0.05).Conclusions:PMΦsCD14 in SAP/SIRS rat model, integrated traditional and western medicine group is obviously lower than SIRS group, traditional medicine group and western medicine group; there is no difference between integrated traditional and western medicine group and control group.This result implies that Da Cheng Qi Tang treatment has inhibit effect on PMΦsCD14 in SAP/SIRS. Integrated traditional and western medicine group is better than simple traditional medicine or western medicine treatment.
