On this basis, to investigate the expression of signal transducer and activator of transcription3(STAT3) and p53 by Immunohistochemistry method and fluorescene Quantitative RT-PCR at mRNA and protein level. The function of the Oxymatrine was Studied that inhibit the proliferation of human colon cancer cells SMMC-7721 actitivities on the adjustment of gene expression and the cell differentiation.The experimental results:Cultured SMMC-7721 in vitro, with the increasing concentration of Oxymatrine and Extension of time , the inhibition rates rise; at the same time, the nucleus shrinkage; cell structure unclear; in the number of apoptotic cells increased; a large number of cell detached, and the SMMC-7721 arrest in G0/G1 treatmented with oxymatrine. Immunohistochemistry,Compared with the control group, the positive expression of p53 protein in SMMC-7721 cell was upregulated, and the expression of STAT3 protein in SMMC-7721 cell were significantly decreased after treated with Oxymatrine by different hours. fluorescene Quantitative RT-PCR,p53 mRNA expression levels was increased, STAT3 mRNA expression levels was decreased, with negative correlation between the expression intensity of p53 and STAT3 in SMMC-7721 after treated with Oxymatrine (P<0.01), Compared with the control group, significant difference (p <0.05).Conclusion:(1) The inhibitory effect of Oxymatrine treated SMMC-7721 in vitro has time- dose-dependence natrue, the most obvious inhibited cell growth of Oxymatrine in the 2.0g /L, 48h better than 24h.(2) Treatment the SMMC-7721 by oxymatrine arrested in G0/G1. can not synthesized the DNA of cell growth required , To inhibited the tumor growth.(3) Oxymatrine can influence p53, STAT3 protein positive expression to inhibit cancer cell growth, with negative correlation, positive expression rate of p53 protein increased, the positive expression rate of STAT3 protein declined. (4) Oxymatrine enable the expression of p53 mRNA of SMMC-7721 rising, STAT3 mRNA expression was reduced, 48h more obvious, 2.0g / L best. |
