| Objective:The purposes of this study were to dynamically measure the levels of Interleukin-17 and Interleukin-23 axis molecules in reflux esophagitis rat model and to assess their effect on the pathological process.Methods:The experiments were performed on 30 male SD rats. The rats were randomly divided into 3 groups:namely, a control group, a sham-operated group and a model group. Reflux esophagitis was induced by adopting partial pyloric ligation plus cardiomyotomy. Four weeks later, pathological changes of esophagus mucous membrane were observed by using HE staining in every groups. IL-17contents in esophageal tissues were measured by ELISA; the expression of IL-23p19 and IL-17 mRNA in esophageal tissues were examined by real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (RT-FQ-PCR) with S YBR Green.Results:Compared with the control group, model group had significantly pathological changes of esophagus mucous membrane. Expression of IL-23p19 and IL17 mRNA in the esophageal tissues of the model group was significantly higher than that of the other 2 groups (P< 0.05); the expression of IL-17 protein was significant higher in the model group than that in the other 2 groups (P< 0.05), with no significant difference found between the namely,a control group and sham-operated group.Conclusion:IL-23/IL-17 axis is involved in the pathogenesis of RE. Part two:Influence of ShuGanHeWeiJiangNi Decoction on expression of interleukin-23 and interleukin-17 in esophageal tissues of rat with reflux esophagitisObjective:To study the influence of shu gan he wei jiang ni Decoction on the expression of IL-23 and IL-17 in esophageal tissues of rat with reflux esophagitis and to study the possible mechanism.Methods:The experiments were performed on 50 male SD rats.The rats were randomly divided into 5 groups:namely,a control group, a sham-operated group, a model group,a shu gan he wei jiang ni Decoction group and a omeprazole group. Reflux esophagitis was induced by adopting partial pyloric ligation plus cardiomyotomy. Shu gan he wei jiang ni Decoction group and omeprazole group were given by gastric perfusion from one week after the operation once daily for 28 days. Pathological changes of esophagus mucous membrane were observed by using HE staining in every groups. IL-17 contents in esophageal tissues were measured by ELISA; the expression of IL-23p19 mRNA in esophageal tissues were examined by real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (RT-FQ-PCR) with SYBR Green.Results:Compared with the model group, Shu gan he wei jiang ni decoction group and omeprazole group had significantly lower histological scores (P< 0.05) Expression of IL-23p19 mRNA and IL-17 protein in the esophageal tissues of the model group was significantly higher than that of control group, sham-operated group and shu gan he wei Jiang ni decoction group (P< 0.05); with no significant difference found between control group and Shu gan he wei jiang ni decoction group.Conclusion:Shu gan he wei jiang ni Decoction can effectively inhibit inflammation in rat with reflux esophagitis through non-selectively inhibiting the expression of IL-23p19and IL-17. |
