Background:Artesunate is recognized as an effective antimalarial, in the clinical treatment of malaria has made a good effect. In recent years, many studies have found that artesunate has anti-tumor effect in vitro. A small amount of literature reported that artesunate can induce apoptosis of leukemia cells in vitro, and this new use of its anti-leukemia provides an experimental basis. The problem of drug resistance of leukemia is getting worse, and the existing anti-leukemia drug has been difficult to meets the need. To develop a new anti-leukemia drug is necessary. If artesunate could be developed as a kind of anti-leukemia drug, it will provide a new option for the treatment of leukemia.Objective:To study the mechanism of artesunate on human acute leukemia primary cells, so as to provide further experimental evidence for the artesunate to be used in clinical.Methods:Selected 10 patients with newly diagnosed or relapsed acute leukemia in patients with primary bone marrow cells were cultured in logarithmic growth phase cells were seeded in 6 well plate. To a final concentration of 50μg/ml,25μg/ml,12.5μg/ml of artesunate affects on the cells. Cells were labeled probe JC-1 fluorescence and fixed. We observed cell morphology in the laser scanning confocal microscope, and calculated the red, green fluorescent cells ratio. So that we can arrive at the proportion of cell death, to explore artesunate induced apoptosis in primary acute leukemia in rats.Result:The results showed that artesunate acts on primary human acute leukemia cells, can induce the decrease of mitochondrial membrane potential, and this effect is dose dependent. Among them, the middle dose group and high dose groups were compared with the low dose group was significant. However, the middle dose group and high dose group showed no significant difference.Conclusion:Studies suggest that artesunate has a good anti-leukemia activity. One of its mechanisms is through reduces the cell line plastochondria membrane potential, then causes in its butcher the environment change to cause the tumour cell death.
