Objective:On the foundation of early research, to study BMSCs and Chinese medicine monomer ICA on rabbit osteoarthritis (OA) model of Osteoarthritis of the knee cartilage protection and restoration of function, by Bone Mesenchymal Stem Cells (BMSCs) and Icariin (ICA) knee injection. with a view to BMSCs and ICA on OA cartilage injury protection of possibility and explore its mechanism,by the oberservation of serum I L-1βand cartilage II collagen and expression of Caspase-3. For application and BMSCs repair of cartilage damage, provide a new entry point and methodology for reference in the study of prevention and treatment of OA.Methods:Through the method of primary cell culture, establish a system to culture rabbits BMSCs,and provide the corresponding concentration of the liquid(BMSCs, ICA and BMSCs farming solution) for subsequent experiments.30 Health New Zealand rabbits, half male and half female, weight 2.0±0.3kg, then classified into fivegroups, A set is blank group, group B is model group, group C is BMSCs joint injection group, group D is ICA joint injection group, group E is ICAand BMSCs farming liquid joint injection groups, each group 6 rabbits. In addition to blank groups are modeled with injection of papain into the left hind limbs knee joint cavity. Through observation and collection blood in auricular vein,to test serum IL-1βcontent, in addition to determine the success of modeling. Group A were given weekly 0.2ml saline joint injection. Group B were given once a week with normal saline 0.2ml intra-articular injection; group C were given a 5×104/ml concentration of BMSCs 0.2ml intra-articular injection every week; group D were given a 0.2mlICA injection every week with a concentration 1×10-5mol/L; group E were given 0.2ml farming liquid intra-articular injection once a week. Intervention of the joint are left knee. After the intervention 4 weeks,we executed for all animals. Anatomy of the left knee, the first general observation of the groups of animals on the surface of the articular cartilage damage, and the left knee cartilage by optical microscopy histomorphological changes; using ELISA to detect IL-1βof content; use Immunohistochemistry assay on the cartilageⅡcollagen, to detect expression of Caspase-3. Results:Through the morphology observation and the identification with differentiation, the cultured cells is a rabbit BMSCs. ICA and BMSCs farming liquid joint injection group, and simple BMSCs joint injection group and simple ICA joint injection group comparison, serum IL-1βcontent and cartilage of Caspase-3 expression has dropped significantly, statistics are significant difference (P<0.05); and ICA and BMSCs farming liquid joint injection group can significantly improve articular cartilage II collagen expression, the rest of the group of cartilage II collagen expression results compared with farming solution group, ICA and BMSCs farming liquid joint injection group has significant statistical significance (P<0.01)。Conclusion:ICA and BMSCs farming liquid joint injection can control rabbit osteoarthritis of cartilage damage. There is an interaction between the two. They can protect the cartilage type-II collagen, and inhibit participation chondrocytes of Caspase-3. Its mechanism of action may be through the ICA to promote BMSCs in the joint to proliferate or suppression the production of cytokine which participate in the apoptosis in chondrocytes, thereby reducing the negative effect of cytokines on cartilage cells and cartilage matrix injury and to promote the BMSCs differentiation into cartilage cells, improve the quality of the cartilage repaired. Its mechanism from the theory of Chinese medicine, is that epimedium can temperature-Yang,expelling wind-damp rheumatism and Qi can unblock the stasis of blood in the first. The final part, BMSCs complement deficiency of the kidney, both of them combined supplement both yin and Yang, encouraged and promoting BMSCs play a positive role.
