Objective: Studies effect of T lymphocyte subsets in anti-CD45RB monoclonal antibody(anti-CD45RBmAb) inducted immune tolerance. The investigate the potential mechanism of T lymphocyte subsets in anti-CD45RB antibody-induced immune tolerance may play a role.Methods:T lymphocytes were enriched from spleen of C57BL/6 mice with immunomagnetic beads. Co-culture ConA with T lymphocyte in presence or absence varying doses of anti-CD45RB mAb for 72 hours in vitro. The proliferations of T lymphocytes were measured by CCK-8 kit. Splenocyte were enrichied from BALB/c mice as stimulates cell, co-culture T lymphocyte with splenocyte in presence or absence of anti-CD45RB mAb for 72 hours in vitro. The hearts from BALB/c mice as donors transplantated into C57BL/6 mice received anti-CD45RB monoclonal antibody The survival rate of receptor mice treated by anti-CD45RB mAb were observed. The peripheral blood, spleen and heart transplantation from reciptient mice were collected in 0,1,3,5,7 days after transplanting. The percentage of Thl (CD3+CD8-IFN-γ+),Th2 (CD3+CD8-IL-4+),Treg (CD4+CD25+CD127-),Ts (CD8+CD28-),Tm (CD4+CD62L-CD44hi) in vitro and vivo were detected by flow cytometry. The levels of IL-2,IL-4,IL-10 and IFN-y of culture supernatant or blood serum were measured by ELISA. Foxp3 mRNA were examined by Real-Time PCR. Allografts were evaluated by pathologic histological.Results:Anti-CD45RBmAb could prolong the survival of allografs. Further more, anti-CD45RB mAb inhibited ConA stimulated the proliferation ability of T lymphocytes.The percentage of Th2,Treg,Ts cells were up-regulated and Thl,Tm cells were down-regulated when compared with control either in vitro or in vivo. The level of IL-2 and IFN-y decreased in experimental group, but the purity of IL-10 and IL-4 were opposited. Foxp3 mRNA also increased in the experimental group. Pathology showed, the number of infiltrating cells in cotrol group was much more than that in experimental group, and also, the extent of pathologic histological was more severe.Conclusion:Anti-CD45RB mAb could prolong the survival of allografs. Anti-CD45RB mAb induced immune tolerance may involve the up-regulation of Th2,Treg and Ts, Thl cells and Tm cells down, the expression of Foxp3 mRNA enhanced.
