| ObjectiveTo investigate the function of ecto-5'-nucleotidase (CD73) in ischemic preconditioning (IP) of rat livers.MethodsAfter the models of sham-operation and hepatic ischemic were established in rats, IP was performed with 4 cycles of 3 minutes'ischemia followed by 3 minutes' reperfusion. After 30 minutes of ischemic reperfusion, the protein expression of CD73 in rat livers at different time point was detected by western blot, HE staining exam hepatic histopathological changes, and serum enzymatic measurement analyse the changes of LDH, AST and ALT.ResultsThe expression of CD73 protein significantly increased after ischemic preconditioning in rat livers. Pharmacological inhibition of CD73 could abolish the function of ischemic preconditioning. Soluble 5'-nucleotidase could protect the liver damnification of ischemic reperfusion.ConclusionCD73 performed important role in the protective mechanism of ischemic preconditioning for rat liver. Soluble 5'-nucleotidase could be a potential therapy for hepatic ischemia. |
