The Levels Of CD3-CD16+CD56+NK Cells In Preeclampsia

ObjectivePreeclampsia is a worldwide leading cause of maternal death and perinatal mortality. It is thought to have the relation with inflammation, dysfunction of the maternal vascular endothelium,and intolerance between maternal and fetus. Generalized dysfunction of the maternal vascular endothelium is thought to be the central event in pathogenesis of preeclampsia.Nature killer cells (NK cells) are not depend on the antigens investigation.They can lyse the tumor cells and infectious cells spontaneously.CD3-CD16+CD56+cells are the representation of NK cells.Our aim is to investigate the alteration of natural killer cells (NK cells) CD3-CD16+CD56+ Level in immunologic interaction site of maternal and fetus (peripheral blood).The objective is to assertain the relationship between NK cells and preeclampsia.MethodsThe NK(CD3-CD16+CD56+) cells in peripheral blood were determined respectively by cytometric analysis.27 patients with Hypertensive Disorder Complicating Pregnancy who had prenatal examinations and delivered in the Obstetrics of the SHENGJING Hospital were selected as study group including 8 patients with light preeclampsia,19 patients with severe preeclampsia;At the same period,15 normal late-pregnant women who matched for maternal age and gestational age with PE Group and 15 healthy non-pregnant women who matched for age volunteered for the study were selected to serve as two control groups.ResultsIn patients with preeclampsia,the percentage of CD3-CD16+CD56+NK cells in peripheral blood is 15.51%±2.49%,it is higher than that in normal late-pregnant women Group (9.97%±1.41%) and healthy non-pregnant women Group (11.11%±1.48%);there were statistical differences(P<0.01). Women in normal late-pregnant women Group had the lower percentage of NK cells in peripheral blood than those in healthy non-pregnant women Group, but there was no statistical difference (P>0.05).ConclusionsThe percentage of CD3-CD16+CD56+NK cell in peripheral blood was advanced which resulted in enhancement of celluar kill activity,suggesting that interface with preeclampsia immune reaction was abnormal activated in maternal-fetal fetal trophoblastic cell turn into iso-self component and the immune balance was broken,which all contribute to preeclampsia。Preeclampsia may be forecasted and given prevention and cure by determination of NK Cells in peripheral blood.
