| Objective:To study the mature state, distribution and quantity of dendritic cells (DCs) in the decidua of women with recurrent spontaneous abortion (RSA) and normal pregnancy, and investigate the role of DCs in the RSA.Methods:We used the EnVision two-step immunohistochemical staining technique to detect the expression of CD83 and CD1a in the decidua of women with RSA (RSA group,30 cases) and normal pregnancy (control group,30 cases). The mature DCs were characterized by the expression of CD83,and the immature DCs were characterized by CD1a. The location and number of DCs in two groups were observed.The number of positive cells within 30 high power fields(10*40) were counted. RSA is defined as three or more consecutive pregnancy losses.Results:1. The location and number of CD83+DCs in the decidua in two groupsThe decidua from two groups was found to contain DCs that stained with the anti-CD83 antibody. Most of the DCs in the decidua from two groups were located close to maternal glandular epithelium and some of these DCs clustered with other.A few of these DCs were present as single cells in the stroma. No difference in the localization of CD83+DCs was found in the decidua between two groups.The number of CD83+DCs in two groups were14.20±2.44/30HPF and 4.77±0.48/30HPF respectively.CD83+DCs were more numerous in the decidua from women with RSA than normal pregnancy.There was significant difference between two groups (t=3.800,p=0.001).2. The location and number of CD1a+DCs in the decidua in two groupsThe decidua from two groups was found to contain DCs that stained with the anti-CD1a antibody. Most of the DCs in the decidua from two groups were located close to maternal glandular epithelium and some of these DCs clustered with other.A few of these DCs were present as single cells in the stroma.No difference in the localization of CD1a+DCs was found in the decidua between two groups. The number of CDla+DCs in two groups were3.97±0.68/30HPF and7.60±1.11/30HPF respectively. CD1a+DCs were more numerous in the decidua from women with normal pregnancy than RSA.There was significant difference between two groups (t=2.786,p =0.008).Conclusion:1. The increase in the number of mature DCs and the decrease in the number of immature DCs are related to RSA.2. DCs may play an important role in the aetiology of RSA. |
