backgrounds and objections:Ovarian cancer is one of the three genital malignant tumor in female, it has beco me a serious threat to women's lives and health. Because of more ovarian cancer hidde n onset and no obvious clinical symptoms early, ovarian cancer is hard to detect,most clinical cases are found lately,which leadto the unsatisfied treatment results and the low er 5-year survival rate. So it is particularly important to improve the accuracy of early diagnosis and identification diagnosis of epithelial ovarian cancer. P-catenin is a bioact ive protein, which is involved in the adhesion connections and cytoskeleton connections between cells and participates in the Wnt signal transduction pathway.It plays an impo rtant role in the occurrence, development, invasion and metastasis of malignant tumor. Previously studies has shown that as the acting part of the Wnt signal transduction pat hway, the over-depositionβ-catenin in cancer cells can transit into the nucleus to prom ote over-expression of oncogenes which can lead to cancer cell proliferation,β-catenin can also activate the transcription of many genes in tumor cells, such as the E-adhesi on protein and MMPs which are related with tumor invasion and metastasis, to decreas e the adhesion capacity of tumor cells, as a result, the blood flow can wash away the cancer cells easily into the invasion and metastasis chain of tumor. FGF-2 is a kind of cell division promoting factor with a variety of biological activitiions. It can promote proliferation and differentiation of a variety of cells including the fibroblasts, FGF-2 ca n also involve in the tumor angiogenesis, which is closely related to tumor occurrence, development, invasion and metastasis. This experiment aims at through the test ofβ-c ateninand FGF-2 on lymphocytes surface of ovarian cancer peripheral venous blood to supplythe indicator of ovarian cancer pathogenesis,diagnosis,treatment and prognosis. Materials and Methods:32 cases of patients with ovarian cancer; 7 cases of patients with borderline ovaria n tumor; 16 cases of patients with benign epithelial ovarian tumor; 7 cases of patients with mature cystic teratoma; 13 cases of patients with ovarian chocolate cyst and 6 cas es of uterine prolapse patients with normal ovarian tissue. Take peripheral venous blood of all patients in the first hospitalization before surgery and isolate the lymphocytes for research, FCM test the percentage of positive expression ofβ-catenin, FGF-2,CD3,CD4 and CD8 on the lymphocytes surface。Results:Through FCM detection, the percentage of positive expression of P-catenin and FG F-2 on the lymphocytes surface of ovarian cancer peripheral venous blood is higher sig nificantly than benign epithelial ovarian tumor, mature cystic teratoma, ovarian chocolat e cyst patients and uterine prolapse patients with normal ovarian tissue (P<0.05);the percentage of positive expression ofβ-catenin and FGF-2 on the lymphocytes surface o f ovarian cancer peripheral venous blood showed no significant difference in mean fluo rescence intensity with borderline ovarian tumor (P>0.05);the percentage of positive ex pression ofβ-catenin on the lymphocytes surface of ovarian cancer peripheral venous bl ood is related with the FGF-2 (P>0.05);the percentage of positive expression ofβ-cate nin and FGF-2 on the lymphocytes surface of ovarian cancer peripheral venous blood i s related with the volume of ascites, histological type and cytological grade(P<0.05);bu t show no significant difference between the groups of patient's age, clinical stage, size of ovarian tumor (P> 0.05); combining the percentage of positive expression of P-cate nin,FGF-2 on the lymphocytes surface and CA125 in serum of ovarian cancer periphera 1 venous blood,we can improve the diagnosis sensitivity and the rate of positive expres sion of ovarian cancer. the percentage of positive expression ofβ-catenin and FGF-2 o n the lymphocytes surface of poorly-differentiated ovarian cancer peripheral venous bloo d is related positively with the positive expression of CD8 (P<0.05); the percentage of positive expression of P-catenin on the lymphocytes surface of well-differentiated ovaria n cancer peripheral venous blood is related negatively with the positive expression of C D4 and CD8 (P<0.05); the percentage of positive expression ofβ-catenin and FGF-2 on the lymphocytes surface of borderline ovarian tumor peripheral venous blood is relat ed positively with the positive expression of CD3 and CD8 (P<0.05).Conclusions and innovationAs the vector, the lymphocytes in peripheral venous blood of patients with ovarian cancer can carryβ-catenin and FGF-2 into blood circulation and lymph circulation, wit h the lymphocytes homingβ-catenin and FGF-2 can reach many tissues, this prompt th at lymphocytes play an important role on development, tumor-associated antigens presen tation,tumor immrne response and lymphocyte homing; The increase positive expression ofβ-catenin and FGF-2 on the lymphocytes surface of ovarian cancer peripheral venous blood prompt that two of them can play an important role on stimulating lymphocytes proliferation, promoting cancer cell proliferation, invasion and metastasis, promoting ang iogenesis and malignant transformation of normal cells; The percentage of positiveexpre ssion ofβ-catenin and FGF-2 on the lymphocytes surface of poorly-differentiated ovaria n cancer peripheral venous blood is related positively with the positive expressionofCD8, this prompt that three of them play a synergistic role in occurrence of ovarian cancer a nd tumor immunity, the increase positive expression of three of them may prompt high malignancy and poor prognosis; The percentage of positive expression of P-catenin on t he lymphocytes surface of well-differentiated ovarian cancer peripheral venousblood is r elated negatively with the positive expression of CD4 and CD8, this prompt thatthree o f them play a antagonism role in occurrence of ovarian cancer and tumor immunity; th e increasing positive expression ofβ-catenin and FGF-2 on the lymphocytes surface of ovarian cancer peripheral venous blood and the expression of CA125 in serum have co mmon trends and complementary, combining of three of them can improve the diagnosi s rate of of ovarian cancer to some extent; The percentage of positive expression ofβ-catenin and FGF-2 on the lymphocytes surface of ovarian cancer peripheral venous blo od is related with the volume of ascites and histological type, it can be used as the pr ognosis judgement and the guidance of biological treatment of ovarian cancer; Reinfuse cultural lymphocytes in vitro,wich carry specific neutralizing antibody or gene mutation ofβ-catenin and FGF-2, may new ideas for tumor treatment.Our experiment use FCM to detect the expression of tumor-associated factors-β-cat enin and FGF-2 on the lymphocytes surface of peripheral venous blood, not only about tumor cells, it can connect the lymphocytes of blood circulation with the development, invasion, metastasis, prognosis and biological treatment of tumor cells, this is the innov ation of our research.
