Background: Epstein-Barr virus, frequently referred to as EBV, is a member of theγ-DNA herpes virus family and one of the most common human viruses. EBV is a common human virus that plays a role in the emergence of nasopharyngeal carcinoma (NPC). NPC is consistently associated with EBV infection,and almost all NPC patients have sustained high levels of serum antibodies against EBV. Prospective clinical and serological surveys suggest that EB virus-specific antibody detected by serological testing is an important mean of early detection of nasopharyngeal carcinoma patients. EBV serology is nowadays to be routinely used in laboratory examinations for diagnosing NPC as well as in the early detection. Detection of serum EBV-VCA/IgA is widely used for screening of nasopharyngeal carcinoma at present. Recent study shows that EBV-NA1/IgA, EBV-Zta/IgA detection in nasopharyngeal carcinoma patients has some value in the early diagnosis of nasopharyngeal carcinoma. Guangxi Autonomous Region is a High-incidence Area of Nasopharyngeal Carcinoma, has a lot of minority nations as well. It is necessary to detect EB virus-specific antibody in different minority ethnic groups and analyze the major risk factors. The experimental evidences are provided for early prevention and reduce infection rates of experimental and theoretical basis.Objective:The aim of this experiment was to assess three enzyme-linked immunosorbent assays (ELISA), namely, VCA-IgA, Zta-IgA and EBNA1-IgA, for their application in serodiagnosis of nasopharyngeal carcinoma (NPC), to investigate what combinations of ELISAs were more efficient, to assess and find out EBV infection, meanwhile, to analyze and investigate major risk factor for EBV infection. Based on the study of EBV infection status of malignant lymphoma in different ethnic groups(Mulam ethnic﹑Maonan ethnic and Han ethnic). Meanwhile, compare the levels of anti-EBV antibodies in different ethnic groups of Guangxi Autonomous Region,and to illustrate the relationship between EBV infection and the ethnic distribution of NPC.Methods: Healthy natives from Luocheng, Guangxi are collect, including 3060, 1497 Mulam nationalities, 963 Maonan nationalities and 600 Han nationalities. Sex, age, smoking and alcohol consumption and other personal information are collected through the form of a questionnaire. The serum levels of VCA-IgA,EBNA1-IgA, Zta-IgA in 3060 healthy adults were tested using ELISA,and presented by the mean ArA value of VCA-IgA,EBNA1-IgA and Zta-IgA. Invest and statistics EBV infection rate in different gender and age stage and relative risk. The serum levels and positive rates of the three antibodies were compared between the three ethnic groups.Result:1. The positive rate of VCA-IgA is 8.40% among the 3060 case invested, Zta-IgA is 7.12% and EBNA1-IgA is 5.03%.2. It have Statistical difference in gender, the positive rate of VCA-IgA, Zta-IgA, EBNA1-IgA in men is higher than women.3. the positive rates of VCA-IgA, Zta-IgA, EBNA1-IgA in different age stage have statistical difference, Statistical analysis showed that: the range of 50 to 59 years old group,the the positive rates of VCA-IgA, Zta-IgA, EBNA1-IgA is significantly higher than other group of years old (P<0. 05), that the middle-aged and old people have higher infection rate.4.the positive rates of VCA-IgA, Zta-IgA, EBNA1-IgA has nothing to do with smoking and alcohol consumption (P>0.05).5.the positive rates of VCA-IgA between Mulam nationality and Maonan nationality were respectively 9.22% and 8.83%, were significantly higher than Han nationality (5.67%, P<0.05). the positive rates of Zta-IgA between Mulam nationality and Maonan nationality were respectively 8.28% and 7.48%, were significantly higher than Han nationality (3,67%, P<0.05).6.The mean ArA values of EBNA1-IgA of all ages was no statistical significance among three ethnic samples (P>0.05). The mean ArA values of both Zta-IgA and VCA-IgA were significantly higher in Mulam nationality than in Han nationality; The mean ArA values of both Zta-IgA and VCA-IgA were significantly higher in Maonan nationality than in Han nationality; The mean ArA values of both Zta-IgA and VCA-IgA was no statistical significance between Mulam nationality and Maonan nationality.Conclusion:1. the positive rates of VCA-IgA, Zta-IgA, EBNA1-IgA among Mulam ethnic, Maonan ethnic and Han ethnic of 3260 case invested were respectively 8.40%, 7.12%, 5.03; males more likely than women infected with EB virus; middle-aged and old (50 to 59-year- old) have the highest EB virus infection rate. EB virus infection has nothing to do with smoking and alcohol consumption.2. Mulam nationality and Maonan nationality are likely to have an elevation of serum IgA antibodies against EBV lytic antigens(Zta-IgA and VCA-IgA), which represents reactivation of EBV latency infection and implies that Mulam nationality and Maonan nationality may have higher risk to develop NPC than Han nationality.
