| Objective:To use modified wuzi yanzong pill to treat kidney-yang deficiency syndrome of rats which was modelled by hydroxyurea, and study on the mechanism of supplying the kidney essence and protecting testicular spermatogenic.Method:①30 health purered male Wistar rats and from 160-190g liveweight were divided randomly into three groups:normal control group, model group and treatment group. Normal group was given saline lml/100g/d as blank group; model group and treatment group were given lml/100g/d hydroxyurea solution to establish animal model of kidney-yang deficiency syndrome; at the same time, treatment group was given modified concentrated solution wuzi yanzong pill with lml per rats in each time at morning and night. Three groups were administrated continuous by gavage for 15 days and were disposed.②Left testis and epididymis were weighed with the analytical balance.③Cryopreserv-ed right testis in liquid nitrogen subzero 80℃and measured testis tissue homogenate Testosterone.④Using laboratory counter to determine density, vitality, activity and deformity of sperm under microscope. lication of radioimmunoassay in determination of serum and testis tissue homogenate of Testosterone.⑥One-Way ANOVA or rank sum test was done with SPSS 16.0 software for laboratory data.Result:①Comparison weight before and after laboratory rats in each group:no difference of weight in three groups was found(P>0.05).②Observation the general condition of rats in each group:there were a series of deficiency of syndromes in model group with back of arch and twist, activity retardation, hair withered and thinning, even pieces of abscisic. Normal control group and treatment group was no such phenomenon.③Comparison weight of Left testis and epididymis in each group:there were no obvious difference between normal control group and treatment group, in contrast to model group there was significant difference (P<0.05).④Comparison density, vitality A-level motility and deformity of sperm in each group:there were no difference between normal control group and treatment group, in contrast to model group there was difference (P<0.05) in density, vitality and deformity of sperm. There were differences among three groups in sperm motility(P<0.05), but mean number of treatment group was close to normal control group which was higher than model group.⑤Serum and testis tissue homogenate of Testosterone in each group:there were no difference between normal control group and treatment group in Serum Testosterone, in contrast to model group there was difference (P<0.05). There were no differences among these three groups in testis tissue homogenate of Testosterone(P>0.05).Conclution:①modified Wuzi yanzong pill could promote the growth of the testis and epididymis and give the foundation for development, production, mature of sperm.②modified Wuzi yanzong pill could increase the density and A-level motility of sperm, decrease deformity rate, raise the chances of fertility in rats.③modified Wuzi yanzong pill had sex hormone-like effects and could promote the secretion of testosterone. |
