Background:Traumatic brain injury is the most common diseases, its mortality and disability are high. Secondary brain injury is one of the factors affecting the prognosis of patients in diseases, and also a hot research. Studies on traumatic brain injury (TBI) have been largely focused on free radicals and overload calcium, etc, less is reported about the brain protective effect of analgesics. This research is on the protective effect of fentanyl on traumatic brain injury in rats.Object:To observe the pathological changes and the expression of NF-κB, NF-κBmRNA on fentanyl treated traumatic injuryed brain in rats.Methord:18 healthy male Wistar rats (weight 220-250g, purchased from the Institution of Experimental Animals of Shanxi Medical University) were randomly divided into 3groups: control group, TBI group, TBI with fentanyl group (n=8). Feeney TBI animal model were made with apparatus, after 120mins, the rats were killed as scheduled, and the samples were collected for HE, IHC, ELISA, RT-PCR. The expressions of NF-κB in brain of the rats were examined by IHC, ELISA. That of NF-κBmRNA were examined by RT-PCR.Result:HE showed that Fentanyl could reduce interstitial edema of cortex cells after TBI. IHC and ELISA showed that the expressions of NF-κB in TBI with fentanyl group is lower than that of the TBI group (P<0.05).The content of NF-κB mRNA were significantly less, comparing with rats in TBI groups (P< 0.05).Conclusions:Fentanyl can effectively inhibit secondary inflammation injury after acute traumatic brain injury. Its molecular mechanism may be involved in inhibiting the expression of NF-κB,and then reducing the inflammatory response after traumatic brain injury,thus reducing the meninges permeability and cerebral edema.
