The present study was carried out in order to investigate the effectiveness of the QiLan SiJunZi Decoction on immune responses to Newcastle Disease Virus(NDV) vaccine in chickens. Chickens vaccinated with La Sota (â… -â…¢) were given QiLan SiJunZi Decoction 1ml at low(0.125 g/ml), middle(0.25 g/ml), high(0.5 g/ml) dosages by oral administration respectively for 10 days. And other two groups(â…£,â…¤)were given normal saline and La Sota vaccine as control. Peripheral blood samples were taken on days 20, 30, 40, 50 and 60 from ten chickens in each group to determine ND-HI antibody titer. Then they were sacrificed and the weights of whole body, thymus, spleen and bursa of Fabricius were weighed for calculation of immune organ index. At the age of 21, 35, 49 days, phagocytosis activity of macrophages were determined with the experiment of carbon particles clearance. Intestine tissues were sampled on days 20, 40, and 60, The IgA secreting cells and SIgA(Secretory IgA)of chicken small Intestine were determined by means of Immunohistochemical examination and enzyme linked immunosorbent assay respectively. At the age of 20, 40, 60 days, five chickens were sacrificed and picked up the small intestine (jejunum). The mRNA expression of IL-2 and IFN-γof jejunum were detected by semi- quantitative RT-PCR assay. The results showed that high dose of QiLan SiJunZi Decoction could significantly enhance the antibody titers(P<0.01), promote the growth index of immune organs(P<0.05), increase the phagocytosis activity of macrophage(P<0.01). The middle and low dose of QiLan SiJunZi Decoction could enhance the antibody titers, index of immune organs, and phagocytosis activity of macrophage differently. QiLan SiJunZi Decoction could increase the numbers of IgA secreting cells and the levels of SIgA antibody significantly. Meanwhile QiLan SiJunZi Decoction could promote the expression of IL-2 mRNA in the whole immune period, but the expression of IFN-γmRNA was only increased in initial stage. In conclusion, our results show that QiLan SiJunZi Decoction had effects on enhancing specific humoral and cellular immune responses to NDV-immunized chicken. |
