Pine-mushroom(Tricholoma Matsutake Sing.), also known as truffles, pine Tan, Tricholoma matsutake, is Basidiomycotina, layer Basidiomycetes, Agaricales, Tricholomataceae and Tricholoma. Its fruitng bodies possesse of hypertrophy, pleasant aroma and very pleasant tasty. It has a high nutritional value and medicinal value. According to the literatures, pine-mushroom can help to improve the health, to nourish kidney and tonic yang, to improve gastrointestine, to regulate the flow of qi to eliminate phlegm, and deworming. Modern medicine study demonstrate that pine-mushroom also possesses of effect of therapy for diabetes and inhibition of cancer cell proliferation. It is named as "King of mushroom" by Japanese.Pine-mushroom contains of polysaccharide, steroid, saponin, anthraquinones, phenols, tannin, alkloid, coumarin, terpenoids, aroma, triterpene, sterol,8 kinds of essential amino acid, vitamin ets.In our paper, we will study on the volatile components, polysaccharide and other constitents of pine-mushroom. The method of steam distillation and GC-MS are used to separate and analyse the volatiles, and 48 kinds of compounds were identified. SephadexG-150 and RP-HPLC were used to purify polysaccharide and determinate its component of the monosaccharides in polysaccharide.6 kinds of component were determinated and identified as D-Glu, D-Gal, D-Man, D-Rbi, D-Xyl, D-Fuc. In this research pine-mushroom was extracted by 90%and 75%ethanol and then was suspended in water and successively partitioned and extracted with petroleum ether, dichloromethane, n-butanol. The portion of the petroleum ether fraction, dichloromethane fraction,n-butanol fraction were separated by silica gel column chromatography, macroporous adsorption resin, SephadexLH-20 column chromatography. 10 compounds were isolated from pine-mushroom on the basis of spectral data (such as, and were identified as 1H-NMR,13C-NMR,DEPT ets), and were identified as ergosta-5,7,22-trine-3β-ol (Ⅰ), ergosta-6,22-diene-5 a,8α-epidioxy-3β-ol (Ⅱ), ergosta-5,22-diene-3β,7β-diol (Ⅲ), ergosta-7,22-diene-3β,5 a,6β-tiol (Ⅳ), ergosta-7,22-diene- 3β-ol(Ⅴ), ergosta-7,22-diene-3β,5a,6a,9a-tetrol (Ⅵ). This is the first time that compoundⅠandⅢhave been isolated from Tricholoma Matsutake Sing.. All of the compounds are the first time that have been isolated from Tricholoma Matsutake Sing. Changbai mountain.China. The pharmacology demonstrated that compoundⅡhas cytoxic effect on cells of U266.U937 and MM1.S.
