| Objective:To explore the role of FEN1 mutations in the pathogenesis of lupus nephritis (LN).Methods:Peripheral blood samples were obtained from 43 LN patients and 26 healthy subjects. Genomic DNA was extracted from peripheral blood utilized DNA extraction kit. FEN1 gene sequences were amplified by PCR using genomic DNA isolated from LN patients and healthy subjects as a template. All PCR products were analyzed by direct DNA sequencing. In order to search for possible mutable site, we compared sequencing results to FEN1 in gene database. Finally, all data were analyzed by chi-square criterion.Result:Through forward direction DNA sequencing, we discovered LN patients have one FEN1 deletion mutation.Conclusion:LN patients have one FEN1 deletion mutation. |
