Study The Expression Of VHL Gene And VHL Protein In Sporadic Pheochromocytomas

Objective VHL gene is a tumor suppressor gene,which situated in 3p25-26 of chromosome. The function and expression changes of VHL gene is related to the pathogenesis of pheochromocytomas. it was found germline mutations in VHL gene, cause pheochromocytomas. The purpose of our experiment is to study the expression of VHLmRNA and pVHL in sporadic pheochromocytomas, to search for their relationship.To findout the differrece between the pheochromocytomas without gene mutation and the pheochromocytomas without gene mutation,and then,approach the clinical significance.Methods Select 40 pheochromocytomas patients in our hospital which has operated and were final diagnosis in pathology from 2007.07 to 2009.06.All the patients hasnot got any radiotherapy and Chem before operation. At the same time, collection normal adrenal gland come from health adult die of accident.(1) By immunohistochemistry, we investigated the expression of pVHL in normal adrenal gland and pheochromocytomas.(2) By In Situ Hybridization, we make use of man-made VHL gene probe investigated the expression of VHL mRNA in normal adrenal gland and pheochromocytomas.(3) Contrast different organizations stationary protective effect on the human leukocyte antigen.Results (1) The expression of pVHL between sporadic pheochromocytoma and normal adrenal medulla has no statistical significance. Through Spearman correlation analysis, we found that the expression between pVHL and HIF-1a,HIF-2awas negative correlation. There were has no relations between HIF-la and HIF-2a.(2) The expression of VHLmRNA between sporadic pheochromocytoma and normal adrenal medulla has statistical significance. Through Spearman correlation analysissporadic pheochromocytoma and normal adrenal medulla, we found that the expression between pVHL and VHLmRNA was positive correlation.(3)compared the antigens protective effect of 4% paraformaldehyde 10% andformaldehyde, P<0.05, the difference was statistically significant. Conclusion (1) There was negative relationship between pVHL and HIF-a in pheochromocytomas, Which has great significance in occurrence and development of pheochromocytoma, and possible become one of the target for the treatment and diagnosis of pheochromocytoma.(2) In sporadic pheochromocytoma and normal adrenal medulla,There was positive relationship between pVHL and VHLmRNA.(3)The antigens protective effect of 4% paraformaldehyde is much better than 10% formaldehyde.
