In recent years, dendrimers are reported as gene delivery vector, among which polyamino amide (PAMAM) is one of the most frequently studied. In this paper, PAMAM G4 and PAMAM G5 were used as DNA vaccine delivery vector. Different amount of PAMAM and plasmid DNA were combined to make a complex, agarose gel retardation assay showed that both PAMAM G4 and G5 can form complex with DNA. The enzyme protection results showed that PAMAM can protect cargo DNA from being degraded.Toxicity is one of the major concerns in application of PAMAM dendrimers as a gene delivery vector. MTT experiment was carried out on different cell lines including HEK 293, HepG2, U251 and A549. PAMAM G5 are more toxic compared to PAMAM G4, at the same time, the toxicity increased with the concentration of PAMAM applied. Apoptosis study showed that PAMAM induced cell death in 24 hours, the amount of cells dead is generation and concentration dependent.Immunization of Balb/c mice was explored, and antibody against the RSV F protein was detected after immunization.
