ObjectiveDetection of programmed cell death-5 (PDCD5) in renal cell carcinoma and normal kidney tissues, and depending on the clinical stage and TNM staging PDCD5 expression, to further explore the expression of renal cell carcinoma PDCD5 change and its role in the development of renal cell carcinoma.Methods一,Tissue collectionExperimental group:Renal cell carcinoma tissue samples taken from May 2004-2009 in June in the Fourth Affiliated Hospital of China Medical University in renal cell carcinoma resected specimens of 43 cases of paraffin, Including 30 males and 13 females, age 37-75 years; According to TNM stage:T1 of 18 cases, T2 of 12 cases, T3-4 in 13 cases;According to pathological stage:I of 15 cases,Ⅱstage 15 cases,Ⅲ-Ⅳin 13 cases, All patients before surgery did not line up, chemotherapy and immune therapy. All the kidney samples were confirmed by pathology.Matched group:Take the adjacent normal renal tissue 20 cases (confirmed by pathology) as normal control.二,MethodsImmunohistochemical SP method detected renal cancer and normal kidney tissue PDCD5 expression.三,statistical analysisComparison of PDCD5 expression in different clinical stage and TNM classification of renal cell carcinoma by Kruskal-Wallis H test, Fisher's exact test and correlation measurement, the analysis process by SPSS 13.0 software, the difference Result1.Normal renal tissue PDCD5 positive staining mostly yellow or brown, cytoplasmic staining was mainly found in renal tubular cells, renal cell carcinoma PDCD5 positive staining mostly light yellow or yellow. PDCD5 in normal renal tissue expression rate was 100%(20/20), while positive expression of renal cell carcinoma was 72.1%(31/43), both significantly different, P<0.05.2. PDCD5 in different clinical staging of renal cell carcinoma, I positive rate of 93.3%(14/15), II of the positive rate was 73.3%(11/15),Ⅲ-Ⅳpositive rate of 46.2%(6/13). The difference was significant (P<0.005). Correlation of different clinical stages in the overall correlation coefficient is-0.526.3. PDCD5 in renal cell carcinoma with different TNM staging, T1 of the positive rate was 94.4%(17/18), T2 of the positive rate was 75.0%(9/12), T3-4 positive rate was 38.5%(5/13). The difference was significant(P<0.005). Correlation of different TNM stages in the overall correlation coefficient is-0.569.Conclusions1. PDCD5 in normal renal tissue was significantly higher than expression in renal cell carcinoma, PDCD5 may be associated with renal cell carcinoma development.2. PDCD5 positive expression of renal cell carcinoma clinical stage and TNM stage was negatively correlated (correlation measurement of the correlation coefficients were-0.526 and-0.569), the higher stages, the positive rate is lower.3.Renal cell carcinoma development may be reduced or missing the PDCD5, PDCD5 in renal cell carcinoma invasion and metastasis may have inhibited the process of malignant transformation in the protective effect. |
