| Objective: Use cortical granule (CG) to evaluate the oocyte cytoplasm maturation, to investigate the relationship between the state of oocyte maturation and the migration and distribution of CG, to know the differences of oocyte cytoplasm maturity in different ovarian response of patients.Method: To collect discarded unfertilized oocytes (GV,MI,MII) from 42 pairs who need assisted reproductive treatment of IVF. According to the ovarian response in women divide to different groups. Using transmission electron microscope(TEM) method and FITC-PNA fluorescence labeling to observed the distribution and migration of CG.Results: The female age in poor ovarian response group were significantly higher than other groups, and the difference was statistically significant (p=0.000). Use electron microscope method and FITC-PNA observed by fluorescent labeling both can be seen as the maturation of oocytes, CG gradually migrated to the cell membrane. Mature state of CG observed fluorescent labeling, with the oocyte maturation rate increased, the fluorescence marked increase in the proportion of grade III (trendχ2=43.828,p=0.000). The fertilization failure of MII oocytes in three different ovarian response were observed in CG fluorescent distribution. The results showed this: in the poor ovarian response group ,CG marked in grade III only 3.33%, lower than the normal ovarian response and ovarian response group, the difference was statistically significant (p <0.05).Conclusion: CG distribution and migration has directly relation with maturation of oocytes , its characteristic can be used to evaluate the maturity of oocyte cytoplasm. FICT-PNA labeled CG in oocytes can be observed by fluorescence microscope distribution, used to assess the maturity of oocyte cytoplasm. This method is more suitable to the clinical observation. Poor ovarian response in patients with failed fertilization oocytes may be have the worse maturity in the cytoplasm. The IVF fertilization failure or low fertilization oocytes in patients useing methods to evaluate the maturity of cytoplasm, can useful to selecte the next cycle of treatment, methods of fertilization and provides a theoretical basis to in vitro culture system. |
