Recombination activating genel(Ragl) plays a critical role in the development and maturation of lymphocytes,and it is related to immune memory functions.It had been proved that deletion of Ragl resulted in a lack of mature functional B and T lymphocytes.Researchers found that Ragl exited not only in immune system,but also in nervous system of mammal,amphibian animals and osteichthyes.The Ragl transcriptions were mostly apparent in regions of the postnatal brain with a high neuronal cell density:the cerebellum and the hippocampal formation,which were relevant to learning and memory function.In this research,we constructed three pairs shRNA targeting Ragl and one-pair independent sequence(negative),packaged with lentiviral vector system,and transferred into the cultured rat hippocampal neuron cells in vitro respectively.We observed by immunofluorescence,assayed for expression of Ragl by real time PCR and Western blot.Lentivirus was rejected into CA3 of the hippocampus in the rat brain stereotaxic apparatus.The expressions of Ragl mRNA were detected by RT-PCR on 7 d,14 d and 28 d after injection.We observed the GFP expression by immunofluorescence,and hippocampal formation by Nissl's staining. Then we chose the 28 d group rats for Morris water maze,we used the place navigation and space exploration.We didn't found experimental group had remarkably disparity compared with the control and negative groups.In long-term potentiation (LTP) research,experimental group had no remarkably disparity compared with the control group.Thus we get the conclusion that the decrease of Ragl expression in hippocampus may not affect rats' spatial learning and memory ability.
