Objectives:To evaluate the effect of Qingreziyinhuoxue recipe on diabetic periodontitis and peripheral blood TNF-αand IL-1βof rats by animal experiment, so as to guide the clinical work theoretically on treating diabetic periodontitis.Methods:10 weeks old Wistar rats were randomly divided into control group, model group and Chinese herbal group. The diabetic periodontitis models were made in model group and Chinese herbal group. Streptozocin(STZ) was injected into the abdomen of the rats(60mg/Kg), blood sugar(>16.7mmol/L) was measured on day 3 after injection to ensure the diabetes mellitus models were set up. The first maxillary molars were ligated with wire ligature for 4 weeks to make periodontitis models. After being gavaged with Chinese herb(Chinese herbal group)/normal saline(control group and model group) for 2 months, blood sugar was detected, then, the blood and the maxilla were collected. The level of TNF-αand IL-1βin peripheral blood was detected by enzyme-linked immunosorbent assay (ELISA). The tissue sections were stained with HE for histological examination:periodontal inflammation was quantified by the inflammatory cell infiltration in gingival regions, whereas periodontal destruction was assessed by the alveolar bone loss in the experimental regions. All the results were analyzed by SPSS11.5 software package with one-way analysis of variance. Results:The blood sugar of rats injected with Streptozocin was above 16.7mmol/L whereas the average blood sugar of rats in control group was 5.56mmol/L. The diabetes mellitus models were set up.4 weeks after ligature, histological examination found that inflammatory cells infiltrated in gingival tissue, the periodontal pockets were deepened, the collagen fibrils were relatively in disorder and osteoclast in the alveolar bone was active. Periodontitis models were made successfully. After being gavaged for 2 months, blood sugar of rats in Chinese herbal group was lower than model group (p<0.05). The amount of inflammatory cell infiltration and the alveolar bone loss were both higher in groups model and Chinese herbal than in control group. The differences were found to be significant between groups control-model, groups model-Chinese herbal, and groups control-Chinese herbal (p<0.05). The levels of TNF-αand IL-1βin model group were both higher than control group and Chinese herbal group. There were statistically significant differences between groups control-model and groups model-Chinese herbal (p<0.05), but no statistically significant difference between Chinese herbal group and control group (p>0.05).Conclusions:The diabetic periodontitis models can be successfully established in Wistar rats by injecting STZ into abdomen and ligating the tooth neck with wire ligature. Qingreziyinhuoxue recipe can improve the blood sugar condition of the diabetic rats, lower the level of TNF-αand IL-1βin peripheral blood. It has therapeutic effect on diabetic periodontitis in rats, but systemic therapy is still needed to cure it.
