Objective:To evaluate the effects ofω-3 polyunsaturated fatty acids with aspirin in periodontitis on experimental perodontitis,and further explore theω-3 polyunsaturated fatty acids on inflammatory response in rats.Methods:animal model of periodontitis was made by using 6 to 8 weeks old male SD rats,48 of them co-ligation with silk thread in the mouth and inoculation of bacteria with high carbohydrate diet,after rat model was established,they are randomly divided into saline group(P group),aspirin group(A group),and eicosapentaenoic acid with aspirin group(EA group).P group,A group,EA group were given different experimental drugs by intraperitoneal injection,animals were sacrificed after 2wk and 4wk drug administration.Obtain samples from rat maxilla,4%paraformaldehyde,HE staining,TRAP staining and TNF-αin immunohistochemistry.Observed indexes include:(1) Clinical index:probing depth(PD);(2) histologic index:HE stained histology,TRAP staining of inflammatory alveolar crest 1/3 broken bones cell counts, immunohistochemical staining of inflammatory alveolar ridge top 1/3 of bone cells,to observe the change of optical density of the TNF-αpositive expression;(3) animals were killed after the blood through the heart,centrifugation,ELISA to detect serum in C-reactive protein level changes.Data by single factor analysis of variance statistically.Results:After administration of intraperitoneal injection EA group can be increased with administration time to reduce the periodontal tissue during periodontal inflammation.After 2 weeks,compared with P group,the expression of TNF-αpositive optical density value in EA group significantly reduced in the inflammatory alveolar crest 1/3 of bone cells,while the A group no such effect;EA group significantly lower CRP levels in peripheral blood serum.After 4 weeks,compared with P group or A group,EA group probing depth improved significantly;and compared with P group or A group,EA group has significantly decrease in inflammatory alveolar crest of osteoclasts per unit length count;while EA group could more significantly reduce TNF-αexpression in the inflammation alveolar crest 1/3 of bone cells.Conclusions:ω-3 polyunsaturated fatty acids with aspirin has the inhibitory effect on inflammatory response in animal experimental periodontitis and the destruction of bone tissue in periodontal,the osteoclasts,cytokine expression of TNF-αand the blood serum CRP levels were significanct repressed.
