[PURPOSE】We study the changes of gene expression of K562 cells treated by Polyphylloside II of Paris polyphylla Smith using the gene expression microarray. We try to find out the differentially expressed genes of K562 cells with treatmented by Polyphylloside II of Paris polyphylla Smith.[METHODS】First, extracted total RNA of control group and Polyphylloside II of Paris polyphylla Smith group and assessed the quality of the RNA sample by 1% agarose gel electrophoresis. Second, used cRNA Fluorescent Linear Amplification Kit to label RNA with Cy3 and Cy5,reversed transcript RNA to synthesize cRNA,label control group with Cy3 and Polyphylloside II group with Cy5,then synthesize fluorescently-labeled cDNA, and purify them. and hybridized the labeled cDNA with gene chip.Third, scan the microarray with Genepix4000b scanner. Last, analyzed the genes with altered expression level by bioinformatics to explore the molecular mechanisms of Polyphylloside II of Paris polyphylla Smith.[RESULTS]There are sixty-eight differentially expressed genes in common by two chips,Three genes are up-regulated, which related to apoptosis-related genes. While sixty-five genes are down-regulated, which related to cell cycle, cell proliferation and differentiation,protein synthesis, metabolism and modification, germ cell formation, signal transduction, immune defense mechanisms, angiogenesis[CONCLUSION] The affection of anti-leukemia of PolyphyllosideⅡof Paris polyphylla Smith may be the result of combined action of a variety of those genes.The cell cycle regulated genes may play pivotal role. |
