ObjectiveTo detect the expression of hGITRL gene in colorectal cancer tissues by real-time fluorescence quantitative PCR and to analyze the the relationship between the expression of hGITRL gene and colorectal cancer occurrence.Methods(1) The pMD19-T-hGITRL plasmid was prepared by PCR and T-A clone.To establish a QRT-PCR method for the detection of GITRL mRNA and to evaluate the linear range, specificity and degree of precision of this method.(2) The expression of GITRL mRNA in colorectal cancer tissues(n=20) was detected by real time quantitative PCR and compared with control expression. And the relationship between the expression of hGITRL gene and colorectal cancer occurrence was analyzed.Results(1) We prepared the standard plasmid including hGITRL gene successfully and drew the standard curve.(2) Real time quantitative PCR results indicated that the expression of GITRL in colorectal cancer tissues increased compared with control. And we analyzed the correlation between GITRL and colorectal cancer occurrence.ConclusionThe QRT-PCR method for the detection of GITRL mRNA was establish successfully. The expression of GITRL in colorectal cancer tissues increased compared with control expression by real time quantitative PCR. Moreover, the correlation analysis with colorectal cancer occurrence and development suggested that it shares positive correlation with severity of colorectal cancer. |
