Comparison Of The Anti-tumor Effects On Human Esophageal Cancer Eca-109 Cells In Vitro Among Three 5-Fu Loaded Nanoparticles

Objective:Comparing the three 5-Fu loaded nanoparticles(5-Fu-PLA-NP, 5-Fu-PLA-O-CMC-NP,5-Fu-O-CMC-NP) on human esophageal cancer Eca-109 cells in vitro inhibition and apoptosis-inducing effect.Methods:Esophageal cancer Eca-109 cells were exposed to 5-Fu-PLA-NP, 5-Fu-PLA-O-CMC-NP,5-Fu-O-CMC-NP and 5-Fu at the doses(mol/l) of 1×10-7,1×10-6,1×10-5 and 1×10-4 for 1,3,5,7,9 and 11 days respectively.MTT assay was used to measure the proliferative inhibitive rates of Eca-109 cells.Flow cytometry(FCM) was used to detect the effect of Eca-109 cells on cell cycle distribution and apoptotic rate.Results:The inhibition ratio of Eca-109 cells increased with increasing of the concentration and time of 5-Fu-PLA-NP,5-Fu-PLA-O-CMC-NP,5-Fu-O-CMC-NP and 5-Fu groups.In MTT test,the inhibition ratio in experimental groups and control group cells exist significance difference(P<0.01).And with increasing of the dose and time,the difference is increasing the 5-Fu group continues to the 7th day,after that the difference no longer to have statistics significance.But the 5-Fu-PLA-NP, 5-Fu-PLA-O-CMC-NP and 5-Fu-O-CMC-NP groups have statistics significance until to the 11th day(P<0.01).Meanwhile,Compared with the same condition of 5-Fu group,they have significant difference from the 7th day.There has no significant difference among the three 5-Fu loaded nanoparticles in the some conditions.The result of flow cytometry demonstrated that 5-Fu-PLA-NP,5-Fu-PLA-O-CMC-NP, 5-Fu-O-CMC-NP and 5-Fu induced Eca-109 cells arrest in S phase and apoptosis with dose and time dependently.5-Fu-PLA-NP,5-Fu-PLA-O-CMC-NP and 5-Fu-O-CMC-NP showed stronger effects than 5-Fu after 7 days,the significant difference has statistics significance.The four agents blocks the cells at S phase and 5-Fu-PLA-NP,5-Fu-PLA-O-CMC-NP,5-Fu-O-CMC-NP were more potential than 5-Fu.There has no significant difference among the three 5-Fu loaded nanoparticles in the some conditions.Conclusion:5-Fu-PLA-NP,5-Fu-PLA-O-CMC-NP and 5-Fu-O-CMC-NP inhibit the growth of human esophageal cancer Eca-109 cell with concentration and time dependently,which may be correlated to cell cycle arrest and cell apoptosis.There are more effective than 5-Fu in the growth inhibition and apoptosis promotion of human esophageal cancer Eca-109 cell after 7 days.It suggests that the three 5-Fu loaded nanoparticals have a sustained release effect.There has no significant difference among the three 5-Fu loaded nanoparticles in the same conditions.
