Experimental Research On The Effect Of Traditional Chinese Medicine On The Thyroid Function And Morphology In Iodine Deficiency Induced Hypothyroidism Wistar Rats

Objective:(1)By observing the effect of different Chinese herbal compound on thyroid function and morphology in hypothyroidism rats caused by iodine deficiency,to explore the pathogenesis of hypothyroidism.(2) By observing the thyroid function and morphology in hypothyroidism rats caused by iodine deficiency,to compare the effect of Tonifying the Middle and Replenishing Qi Decoction,Tonifying the Middle and Replenishing Qi Decoction combined with small doses of L-T₄ and routine dose of L-T₄.Material and method:According to the purpose,the experiment was carried out of two sets:(1) A search for the pathogenesis of primary hypothyroidism.(2) The effect of Tonifying the Middle and Replenishing Qi Decoction(补中益气汤)combined with small doses of L-T₄ on thyroid function and morphology in hypothyroidism rats caused by iodine deficiency.Wistar rats were divided randomly into two groups:normal group,the experimental group.Normal group was fed normally.Hypothyroidism models were made with low iodine diet and 1%sodium per chlorate for 19 days,double-distilled water(DDW) for 2 days and then divided randomly into:Model group,Tonifying the Middle and Replenishing Qi Decoction group(补中益气汤组),Rambling Powder group(逍遥散组),The kidney- Qi Bolus group(金匮肾气丸组),Tonifying the Middle and Replenishing Qi Decoction combined with small doses of L-T₄ group(补中益气汤联合小剂量L-T₄组),and routine dose of L-T₄ group(常规剂量L-T₄组).Two sets of experiments shared the normal group,model group,and Tonifying the Middle and Replenishing Qi Decoction group.Treatment factors were given correspondingly.90 days later,Serum TT₃ and TT₄ was measured with RIA and serum TSH with IRMA,The thyroid morphology was observed under microscope.Results:1:Comparison among Tonifying the Middle and Replenishing Qi Decoction group, Rambling Powder group and the kidney-Qi Bolus group.(1)TT₃:The value of Tonifying the Middle and Replenishing Qi Decoction group was obviously lower than normal group,higher than model group,higher than the other two,it has markedly discrepancy when compared with Rambling Powder group,but did not when compared with the kidney-Qi Bolus group.The value of Rambling Powder group and The kidney-Qi Bolus group were obviously lower than normal group,but P＞0.05. (2)TT₄:The value of the other four groups were markedly lower than normal group, but did not have markedly discrepancy when compared with each other.(3)TSH: The value of Tonifying the Middle and Replenishing Qi Decoction group was obviously higher than normal group,lower than model group,lower than the other two,it has markedly discrepancy when compared with Rambling Powder group and The kidney-Qi Bolus group.The value of Rambling Powder group and The kidney-Qi Bolus group were higher than normal group.(4) Relative weight of hypothyroid: The value of Tonifying the Middle and Replenishing Qi Decoction group was obviously higher than normal group,lower than model group,lower than the other two,but did not have markedly discrepancy when compared with Rambling Powder group and The kidney-Qi Bolus group.The value of Rambling Powder group and the kidney-Qi Bolus group were obviously higher than normal group.(5) Morphology change:Different factors can recover the morphology at different level, Tonifying the Middle and Replenishing Qi Decoction was better than the other two.2:Comparison among Tonifying the Middle and Replenishing Qi Decoction group, Tonifying the Middle and Replenishing Qi Decoction combined with small doses of L-T₄ group,and Routine dose of L-T₄ group.(1)TT₃:The value of^·-Tonifying the Middle and Replenishing Qi Decoction combined with little dose of L-T₄ group was obviously lower than normal group(p=0.55),higher than model group(p=0.002),higher than the other two groups,it has markedly discrepancy when compared with Tonifying the Middle and Replenishing Qi Decoction Group(p=0.034),but did not when compared with Routine dose of L-T₄ group(p=0.065).The value of Tonifying the Middle and Replenishing Qi Decoction Group and Routine dose of L-T₄ group were obviously lower than normal group,(p=0.008 and 0.017).(2)TT₄:The value of Tonifying the Middle and Replenishing Qi Decoction combined with little dose of L-T₄ group was markedly lower than normal group(p=0.000),higher than model group(p=0.445),but did not have markedly discrepancy when compared with the other groups(p=0.942 and 0.96).The value of the other two groups were lower than normal group(P=0.000).(3)TSH:The value of Tonifying the Middle and Replenishing Qi Decoction combined with little dose of L-T₄ group was higher than normal group(p=0.276),lower than model group(p=0.004),lower than the other two,it has markedly discrepancy when compared with ronifying the Middle and Replenishing Qi Decoction group(p=0.023) but did when compared with Routine dose of L-T₄ group(p=0.169).The value of Tonifying the Middle and Replenishing Qi Decoction group and Routine dose of L-T₄ group were higher than normal group(p=0.001and 0.018).(4) Relative weight of hypothyroid:The value of Tonifying the Middle and Replenishing Qi Decoction combined with little dose of L-T₄ group was obviously higher than normal group(p=0.332),lower than model group(p=0.001),lower than the other two,but did not have markedly discrepancy when compared with the other two groups(p=0.072and 0.132).The value of Tonifying the Middle and Replenishing Qi Decoction group and Routine dose of L-T₄ group were obviously higher than normal group(p=0.008and 0.02).(5) Morphology change:Different factors can recover the morphology at different level,Tonifying the Middle and Replenishing Qi Decoction combined with little dose of L-T₄ was better than the other two.Conclusion: 1:Tonifying the Middle and Replenishing Qi Decoction can improve the function and morphology in hypothyroidism Wistar rats due to iodine deficiency,so we confer that the weakness of Qi of spleen has played a significant role in primary hypothyroidism.2:Tonifying the Middle and Replenishing Qi Decoction combined with little dose of L-T₄ was superior to simplex Tonifying the Middle and Replenishing Qi Decoction and simplex routine dose of L-T₄ in improving the function and morphology in hypothyroidism Wistar rats due to iodine deficiency.
