Effect And Mechanism Of Purification In Bombys Batryticatus On Antithrombosis

Objective: To investigate the effect of purified liqiud in bombys batryticatus(PlIBB) on arterial thrombus and venous thrombus, in order to afford a new approach for seeking an effective antithrombosis medicine from Chinese medicine.Methods: The rabbits and rats were randomized into five groups: blank group,control group and 3 doses PLIBB group (9mg/kg or 18mg/kg or 36mg/kg PLIBB).The effects of PLIBB on antithrombosis were investigated in the rabbits thrombus models established by being damnified the common carotid artery with H₂O₂. The effects of PLIBB in different doses were investigated in the venous thrombus models by ligated the abdomial vena of rats.Results: 1 .The effects of arterial thrombus: (1) PLIBB could alleviate the weight of thrombus; (2) Compared with model group, 3 doses PLIBB group could increase the tissue-tye plasminogen activater (tPA) and antithrombin III (AT-III) activity (P<0.01) , and inhibit the plasminogen activator inhibitor (PAI) activity markely (P<0.01) ,so that the content 6—keto—PGF_1α was increased significantly (P<0.05) and the content of TXB₂ was reduced markedly (P<0.01) in blood plasma; (3)Compared with the modle group,PLIBB signifiantly increased the content of tissue factor pathway inhibitor (TFPI ) and reduced tissue factor (TF) content (P<0.01).2. .The effects of venous thrombus: (1) The PLIBB could inhibit experimental venous thrombosis in rats. Compaqred with model group, the thrombus weight of 3 doses PLIBB group was signifiantly alleviated(P<0.01). (2) Compaqred with model group, PLIBB could decrease the content of fibrinogen (Fbg)and plasminogen(PLg) (P<0.01),so that increase the activity of tPA and AT- III,inhibit the activity of PAI(P<0.01). Meanwhile,the activated partial thromboplastin time (APTT),prothrombin time (PT) and throbmin time (TT) were prolonged obviously(P<0.01).Conclusions: PLIBB holds significant effects on antithrombosis, which may be brought by inhibiting the activity of platelet and slowing down of activating plasminogen into plasmin. PLIBB increases the activity of fibinolytic system through decreased the activity of blood coagulating factors,so that inhibits venous thrombosis.
