Objective: To explore the capability of adipose tissue-derived stem cells of rat differentiating into osteoblasts and adipocytes in vitro. To study the effect of different concentrations of Vancomycin on proliferation and osteogenic differentiation of adipose tissue-derived stem cells of rat in vitro.Methods: ADSCs isolated from rats were cultured in adipogenic induction medium or osteogenic induction medium with different concentrations of vancomycin. Adipogenic differentiation was tested by oil red. MTT method was used to observe the effect of Vancomycin on cell proliferation. The activity of ALP in ADSCs was measured and calcium tubercle was observed to evaluate the osteogenic differentiation effect of different concentrations of Vancomycin on ADSCs.Results: ADSCs can grow in different concentrations of Vancomycin. Vancomycin doesn't influence cells proliferation at concentrations≤800μg/ ml. Vancomycin inhibit cells proliferation at concentrations≥1000μg/ ml. Vancomycin don't influence the activity of ALP at concentrations≤1000μg/ ml. After adipogenic and osteogenic induction,the stainings of oil red,alkaline Phosphatase and von Kossa were pisitive.Conclusion: ADSCs can be isolated from rat adipose tissue and have the potential to differentiate into adipogenic and osteogenic lineage. Vancomycin has little effect on ADSCs proliferation and osteogenic differentiation. It is a good choice to prevent and cure skeletal infection.
