BACKGROUD AND OBJECTIVESEstrogen is involved in the regulation of a wide array of biological processes. As a transcription factor, the classical estrogen effect is through ERα, ERβand estrogen receptor associated proteinγ. unlike the classical estrogen nuclear receptors, an orphan membrane estrogen receptor GPR30 work by the second messenger system in cells, with high binding affinities with antiestrogen. The studies indicate GPR30 participate in the regulation of cell proliferation, cycle and apoptosis. As a hormone sensitive tissue, estrogen is not only to promote the development of mammary gland but also stimulate the growth of breast cancer. The aim of this study was to analyze the correlation between GPR30 and the cell proliferation by blocking the expression of GPR30 in breast cancer cell SKBR3.METHODSThe expression of GPR30 was blocked by siRNA, and cell proliferation was interfered with 17β-E2, EGF and SB202190. GPR30 mRNA was determined by RT-PCR and the cell proliferation with MTT assay.RESULTSTwo recombined plasmids pshRNAGPR301 and pshRNAGPR302 were constructed successfully, and the transfection was performed with GenEscortTMⅠin breast cancer cell SKBR3. The expression of GPR30 mRNA was decreased to 64.25% and 54.24% in contrast to normal cells. No changes were found in the expression of GPR30 after treatment with 17β-E2, EGF and SB202190 in 1hour and 24hours detected by RT-PCR. 17β-E2 did not affect cell proliferation. Unlike EGF′stimulation, SB202190 suppressed cell proliferation. Similar findings occurred in SKBR3-pshRNAGPR302.CONCLUSIONThere was no significant correlation between GPR30 and the proliferation of breast cancer cells in vitro.
