Objective To differentiate the endothelial progenitor cells(EPCs) and discuss their biology characteristic, study the effect of stem cell transplantation on the chronic deep venous thrombosis, thereby to create a new method for the therapy of the chronic deep venous thrombosis.Methods 1. BMMNCs were isolated from rat bone marrow by Ficoll and cultured with EGM-2MV medium cultivation. MTT assay were used to draw growth curve of cells, and assess the growth character of the EPCs after isolated 14 days. 2. Experimental rat model of chronic deep vein thrombosis were obtained by partial ligation of inferior vena cava and intravenous injection the thrombosin. The rats were randomly divided into three groups: A(n=25), EPCs group, 1ml 106 EPCs transplantation; B(n=25), EGM-2MV medium group, 1ml EGM-2MV medium transplantation; C(n=25), control group, without any treatment. 3. After transplantation, immunohistochemical staining was performed to detect recanalization of the inferior vena cava. Western Blotting of inferior vena cava thrombosis was used to detect VEGF,bFGF protein expression changes. 4. SPSS13.0 software was used for analysis.Results The rat bone marrow EPCs were islated successfully in vitro.The rat model of chronic deep vein thrombosis was successfully set up. The recanalization capillary density were significantly higher in group A than that in group B,C(P<0.05). The neovascularization were identified by immunohisto-chemical staining of vWF antibody, that were compose of endothelial cells.VEGF,bFGF protein were significantly increased in group A than group B and C.Conclusions EPCs were the precursor cell of endothelial cells, they have improve potentiality to promote angiogenesis and can accelerate organization and recanalization of vein thrombus after transplantation.
