Transhepatic Arterial Chemoembolization Combined With Recombinant Human Endostatin Via Hepatic Artery For The Treatment Of Rabbit VX2 Hepatoma

Objective:To analyze inhibitory effect and safety on rabbit VX2 hepatoma by Transhepatic arterial chemoembolization(TACE) combined with re-endostatin via hepatic artery.Materials and Methods:The rabbit VX2 hepatoma models were randomly divided into 3 groups. Group A was treated with physiological saline via hepatic artery. Group B was treated with TACE only. Group C was treated with TACE combined with re-endostatin via hepatic artery. Following treatment, the tumor volume(according to the formula:V= ab2/2)and growth rate were measured and calculated on the 1th day,the 4th day and the 7th day after treatment respectively. The metastatic rate and microvessel density( MVD)value of the tumors were also measured on the7th day after treatment respectively. All the animals were explored the ALT and UREA before the operation and on the 1th day,the 4th day and the 7th day after treatment respectively. The data w statistically analyzed by SPSS13.0 statistical bag.Results:1. After the treatment, the majority of experimental animals in group B and group C decreased their appetite apparently. Most of them were in depression and drowsiness and moved litt. One animal was dead in group B and C respectively. One animal's incision disunioned in group C. There were no animals died in group A. ALT level rose apparently on the 1th day and the 4th day after the treatment in group B and group C. Compared with the control group(group A),the differences were statistically significan(tp<0.05).The ALT level was the highest at the first day after the treatment. It showed that the ALT level declined on the 4th day and the 7th day. There was no statistical significance between group B and C. The ALT level was stable before and after the treatment in group A. There were no statistical significance of UREA levels between each group.2. MVD in the group C was more lower than the other groups, and the differences between them were statistically significant(p<0.01). MVD in the group B was higher than group A,but the difference between them was not statistically significant.3. On the 7th day after treatment ,the tumor growth rate of group B and C were slower than the control group(p<0.05).Upon the therapeutic effect of the above mentioned , group C was the best and the tumor volume growth rate was the slowest.4. According to the PET/CT examination on 7th day after treatment, metastatic rate in each group was as follows: group A=80.0%(8/10), group B=70.0%(7/10), group C=50.0%(5/10). The differences were not statistically significant (P=0.133), The most common site of metastasis were liver (14cases), lymph nodes (9 cases), lung (6 cases) and bone (2 cases).Conclusion:The short-term effect on rabbit VX2 hepatoma of TACE combined with re-endostatin via hepatic artery infusion is superior to that of other two groups in suppressing tumor angiogenesis and the growth rate, and re-endostatin causes no more serious kidney and liver dysfunction .
