| The thesis consists of two parts. The first Part is about the chemistry constituents study on the daphniphyllum calycinum Benth. alkaloids. The second part brings forth the study on the daphniphyllum calycinum Benth. alkaloids acticity of anticholinesterase.Daphniphyllum calycinum Benth. is one of the genus Daphniphyllum (Daphniphyllaceae). D.calycinum Benth. is a native evergreen shrub in Southern China. Its leaves and seeds are used in folk medicine to treat several symptoms, including pyretic, inflammation and influenza. While in clinical practice it is mainly used to treat gastroenteritis, dyspepsia and eczema. In search for structurally unique and biogenetically interesting of Daphniphyllum alkaloids, the alkaloids in the seeds and leaves of D.calycinum Benth. collected from GuangXi Province of China were isolated through silica gel chromatograph, reverse-phase RP-18, MCI, gel Sephadex LH-20 and Toyopearl HW-40C. Then 15 daphniphyllum calycinum Benth. alkaloids were isolated in which 12 complicated compounds were established by spectral methods as IR, UV, MS, NMR. Among them, caldaphnidines G-H were two kind of new alkaloids. The other alkaloids were known as calycicine A, daphnezomine L-M, daphnioldhanin G, methyl homosecodaphniphyllate, daphniyunine A, calyciphylline A, calyciphylline B, deoxycalyciphylline B and deoxyisocalyciphylline B. In this study, 2D NMR techniques (~1H-~1H COSY, HMQC, HMBC and NOESY) were used to establish the stereochemistry of these two new alkaloids. The incorrect four structures of the daphnioldhanin D-G previously reported were revised by a single-crystal X-ray diffraction study. Besides, the author also made testing research on the acticity of anticholinesterase of these alkaloids. Though not good biological activities of alkaloids were found, it still will give help to the biological activity of daphniphyllum calycinum Benth.alkaloids study. |
