| Objectives: To investigate the preventive effect of dFMGEN on the damage of hyperglycemic retinal endothelial cells in vitro and provide a new clue And theory basis for developing a drug of preventing and curing diabetic retinopathy(DR) in the future.Methods: Simian retinal endothelial cells incubated with high glucose levels for 72h in vitro, cell function was measured by LDH assay,MTT chromatometry and cell apoptosis was determined by flow cytometry respectively. Experiment groups were divided as follows: (1) blank control group;(2) high glucose damage group;(3) DMSO control group;(4)lead compound group;(5) 7-Difluoromethylyl-5,4'-dimethoxy genistein group;(5) positive medicine control group. The RF-6A cells were pretreated with either the vehicle solvent(DMSO),genistein, dFMGEN and VitB1 for 30 min before exposed to 80mmol/L glucose for 72h. Cell growth and multiplication activity were measured by MTT assay. Cellular injury was estimated by measuring the lactate dehydrogenase(LDH) release. Cell apoptosis was determined by flow cytometry with propidium iodide(PI)staining.Resurlts: simian retinal endothelial cells treated with high glucose levels (80mmol/L) for 72h obviously increased LDH release and inhibitory rate.Cells were incubated with different concentration of dFMGEN before being injured by high glucose levels (80mmol/L).After endothelial cells were pretreated with dFMGEN, experiment results showed: dFMGEN can improve cell growth and proliferation in a concentration dependent manner, reduce the release of LDH, suppress the high glucose levels -induced apoptosis of endothelial cell .1,After 72 hours with different concentrations of dFMGEN (0.1,0.3,1,3,10μmol/L)and genistein(100μmol/L),VitB1(100μmol/L),the inhibitory rates of which were 52.22%,49.02%,46.78%, 45.2%,27.26%,29.11%,30.1% respectively through MTT assay. There were significant differences between experimental groups except concentrations of dFMGEN (0.1,0.3,1μmol/L)(P<0.05).2,After 72 hours with different concentrations of dFMGEN (0.1,0.3,1,3,10μmol/L)and genistein(100μmol/L),VitB1(100μmol/L), the release of LDH of dFMGEN (10,3μmol/L) were significant differences between the high glucose levels -induced groups(P<0.01).3,the apoptosis of endothelial cell were 45.8±2.9%,41.5±2.57%,34.4±5.31%,20.2±%,9.3±1.3%,12.8±4.43%,24±4.74% of dFMGEN (0.1,0.3,1,3,10μmol/L) and genistein,VitB1. dFMGEN (10μmol/L) group is the most effective article among test articles.Conclusion: dFMGEN is more protective activity of vascular endothelial cell injury by high glucose levels in vitro than lead compound- genistein. The protective were increased in dose-dependent ways. |
