Effect Of Matrine On The Proliferation Of Human Corneal FibroBlasts In Vitro

Objective To investigate the effect of Matrine on cell proliferation inhibition and its potential mechanism of human corneal fibroblasts(HCFs) in vitro.Methods Purity of the isolated and cultured of HCFs, observe the growth characteristics of HCFs used trypanblau and MTT assay. HCFs was treated with six concentrations of Mat, set up positive control group, blank control group and solvent control group. Cell proliferation was evaluated by MTT assay in 24h,48h and 72h, then calculate inhibition ratio and IC50. Detect the expression ofâ… andâ…¢type collagen protein mRNA, evaluate the effect on seasoning and dose.Results1. MTT assay indicated that Mat significantly suppressed proliferation of HCFs in dose-dependant and time-dependant positive trend with blank control. Meanwhile, Mat have the obviously stronger inhibitory action than DXM with a same concentration. The IC50 is 125.20 mg/L(24h),80.90 mg/L(48h) and 60.92mg/L(72h).2. RT-PCR indicated that, Mat inhibit HCFs expressing Colâ… in dose-dependant and time-dependant positive trend with blank control, merely high concentration(160mg/L) and long action time(48h) have statistical significance. There is nonsignificance between Mat and DXM in the concentration 160mg/L.3. RT-PCR indicated that, Mat inhibit HCFs expressing Colâ…¢in dose-dependant and time-dependant positive trend with blank control, all group have statistical significance contrasted blank control except 20mg/L. There is significance between Mat and DXM in 160mg/L.Conclusion1. Mat significantly suppressed proliferation of HCFs in dose-dependant and time-dependant positive trend, and 160mg/L Mat have the obviously stronger inhibitory action than DXM with a same concentration.2. Mat may be an efficiently, the poisonous and side effect smaller new medicine to treat haze post refractive surgery ,which can prevent or reduce complications.
