PurposeTo study the expression of toll-like receptor 9 mRNA on peripheral blood and HSV levels in serum in patiants with herpes simplex.Methods287 samples of patients with herpes simplex were accepted.The levels of HSV1 type IgG,HSV1-type IgM,HSV2-type IgG and HSV2-IgM of type by using ELASA in serum.We selected 50 the HSV-positive samples,the HSV-negative 10 cases as a control group.And expressions of TLR9 mRNA on peripheral blood,were detected by using real-time quantitative RT-PCR method.ResultsThe seropositivity rate of HSV(HSV 1 and/or HSV2) was 40.1%(115/287) in 287 patients accepted with herpes simplex.The seropositivity rate of HSV1 was 35.2% (101/287),and HSV2 was 7.3%(21/287);There were 111 samples(38.7%) with IgG antibodies(HSV 1 and/or HSV2)-positive and 4 samples(1.4%) with IgM antibodies (HSV 1 and/or HSV2)-positive.The positivity rate of HSV1-IgG is 34.8%(100/287) in 287 of patients with herpes simplex.And the positivity rate of HSV2-IgG is 6.3%(18/287).The positivity rate of HSV1-IgM and HSV2-IgM is 0.3%(1/287) and 1.0%(3/287),respectively.HSV1-and HSV2- IgG antibodies were both positive in 7 cases.The expressions of TLR9 mRNA were detected with the Real-Time RT-PCR in HSV-positive group and the control group.The relatived expression levels of TLR9 mRNA with the 2-△△Ct values to indicate.The results showed that,HSV-positive group of TLR 9 mRNA expression level of 0.14±0.22,HSV-negative control group, expression level of 0.87±0.23,the difference was statistically significant(P<0.05). The HSV-IgG positive group was divided into HSV1-IgG,HSV2-IgG,as well as HSV1,2-IgG were positive for a total of three groups to compare their relative expression levels of TLR9 mRNA were 0.37±0.13,0.49±0.22 and 0.44±0.09, expression compared with the control group were significantly different(P<0.05).ConclusionIn this study,the expressions of TLR9 mRNA were detect in peripheral blood of HSV-positive patients by quantitative Real-Time RT-PCR method,and were compared with the control group.We found that expression of TLR9 mRNA expression levels of HSV-positive group were significantly higher than the control group.TLR9 plays an important role in the pathogenesis of herpes simple.
