| Nonneoplastic epithelial disorders of vulva is a chronic disease when vulva skin or mucous tissue was denaturized and pigment changed.The disease includes vulva epithelium scalelike hyperplasia,vulva moss cirrhosis and other vulva skin disease, which almost cause impossible oppressive severe itching and influence the patient living badly.The etiological factor of the disease is undefinable,but maybe corelate with chronic damage,hypersusceptibility,parcel malnutrition,metabolic disturbance,immunological diseaseVulva severe itching is a common skin feeling,the major symptom of much systemic disease and skin desease It may be induced by machine or heat incitation, chemical mediator and takes place in central nervous system not depends on external incitation.The results of neurophysiology study has suggested that nerve conduction path takes part in pruritus.The medium involved in pruritus includes neuropeptide substance P(substance P,SP ) and mast cells.Mast cells participates in pathologic process ofâ… -model allergic reaction and much skin diseases,which was brought to important fucnction in immunoreaction and inflammatory reaction by releasing its kernel and matter.SP is the messenger of interaction between nerve and mast cells,which is interrelated with some skin diseasesIn this paper,the organizations of vulva nonneoplastic epithelial disorders in 53 medical case were studied by immunohistochemical method,detecting the expression of SP and mast cells in normal vulva skin,vulva scalelike hyperplasia and vulva cirrhosis moss.The role of mast cells and SP in nonneoplastic epithelial disorders of vulva was studied,and a single step was moved forward to the knowledge of pathology of vulva nonneoplastic epithelial disorders.ExperimentalThe number of mast cells and the expression level of substance P were detected in in nonneoplastic epithelial disorders of vulva by immunohistochemical technique.Material and measurements1.The subject investigatedPathologic specimen whose ages rang forty three years old to senventy years old was got from The first hospital of China Medical University during august and december in 2008.It includes ten instances of nerve normal skins,twenty instances of vulva epithelium scalelike hyperplasia,twenty three instances of vulva moss cirrhosis(ten early period instances,thirteen late stage instances )2.Material and measurementsAntihuman mouse SP cloning vector and mast cells trypsinogen monoclonal antibody were obtained from Maixin Fujian and Maixin US,respectively.High sensitivity s-p activator(mouse/rappit ) and Diaminobenzadine(DAB) were got from Beijing Zhongshan biotechnology Company Limited(Co.Ltd.),3%(V/V) hydrogen peroxide,0.05 M phosphate buffer,10%sheep blood serum,EDTA, dimethylbenzidine(DBA) and haematoxylin were provided by pathology department of The first hosptical of China Medical University.Measurements:pathological section cutter,microwave oven,hatcher, microscope,object carrior,cover thin slice3.Immunohistochemical method(SP)Wax stone of samples was cut into 4μm slices in a row.one slices was stained by HE,and the histology diagnosis differentiation was determined.The expression of mast cells and SP on vulva nonneoplastic epithelial disorders were detected,accord to instruction of reagent box.Then was colourated by DAB.All the samples was stained by immunohistochemistry method affter restoration in microwave oven. Positve cells group was used as positve control and PSB instead of Nestin as negative control.ResultsCompared with normal skins(9.36±3.82),the number of mast cells in squamous hyperplasia of vulva(18.44±2.69),lichen sclerosus(22.25±3.02) was significantly higher(P<0.05);The number of mast cells in the early stage (16.13±2.64 ) was significantly lower than that of in the late stage(20.12±3.67 ) of lichen sclerosus(P<0.05);Compared with squamous hyperplasia of vulva (15.35±2.68 ),the number of substance P in normal skins of vulva(6.46±2.02), lichen sclerosus(8.12±3.52) was significantly lower(P<0.05);The number of mast cells in the early stage(5.62±2.57 )was significantly lower than that of in the late stage (8.65±2.13) of lichen sclerosus(P>0.05)。ConclusionMast cells might play a certain role in nonneoplastic epithelial disorders of vulva; Substance P might play a certain role in squamous hyperplasia of vulva。... |
