Pueraria Lobata Willd is a plant of perennial legume Fujiki deciduous.Its root is called Kudzu Root.It is announced for the medicine,the food dual purpose plant. There are more than a dozen varieties Kudzu Root in my country.Kudzu Root distributed in most of China's provinces and autonomous regions,and it is rich resources.Kudzu Root has been used in medicines and food.The main active ingredient of Kudzu Root is puerarin flavonoids and puerarin.Puerarin is paid more and more attention increasingly for its good therapeutic effect to Brain obstruction, and it has been widely used in the clinic.The experimental study of the extraction,purification and antioxidation property of puerarin flavonoids and puerarin is given in this paper,and then the technology parameters in process were optimized for deciding the best technology.And the application in the food of pueraria lobata has been discussed.The results show that the optimum extraction process of extraction of puerarin flavonoids from pueraria lobata ohwi with ethanol refluxing process is that the ethanol was used as a solvent,extraction temperature 80℃,alcohol concentration of 70 percent,from time 3 h,extract three times and solid-liquid ratio is 1:10.The purity of puerarin flavonoids is 29.31%,and the extraction rate of puerarin flavonoids is 22.9%.The optimum extraction process of extraction of puerarin flavones from pueraria lobata ohwi with ultrasonic-assisted extraction is that the ethanol was used as a solvent,extraction temperature 50℃,alcohol concentration of 70 percent,from time 30 min,extract three times and solid-liquid ratio is 1:10,the ultrasonic power is 300 W.The purity of puerarin flavonoids is 33.041%,and the extraction rate of puerarin flavonoids is 24.1%.Puerarin flavones are extracted by Ultrasonic extraction method is more purity than ethanol extraction method.But in the production costs and inputs, puerarin flavonoids is extracted by ethanol extraction method is save more,so it is apply to small and medium enterprises,and puerarin flavonoids is extracted by ultrasonic extraction is apply to large enterprises.When puerarin was separated,ethyl acetate,butanol was used to extract puerarin flavonoids one by one,then the puerarin-purification was getted from n-butanol extract of the puerarin flavonoids extract by using crystal-crystal method and a column chromatography separation method.HPLC was used in the determination of puerarin in puerarin-purification.The results show that the purity of puerarin-purification isolated by silica-gel column chromatography is 91.1%,and the extraction rate of puerarin-purification is 1.19%; the purity of puerarin-purification isolated by AB-8 macroporous resin column chromatographic is 97.3%,and the extraction rate of puerarin-purification is 1.42%. To consider the safety of food,it was more appropriate that puerarin was separated by silica-gel column chromatography.The antioxidant activity of puerarin flavonoids and puerarin were determined by chemical luminometry and DPPH free radical assay,and are compared respectively on antioxidant effect of propolis flavone,VE,BHT.And it was used to evaluate the antioxidant effect.The results showed that Significant clearing efects of puerarin flavonoids and puerarin were observed On DPPH free radicals,hydroxyl radical (·OH),superoxide anions(O2-),and its antioxidant effect is stronger than BHT,so puerarin flavonoids and puerarin is a anti-oxidants of moderate intensity.Puerarin flavonoids samples of experimental preparation are developed of radix puerariae beverage.The optimum formula of the product was determined as follows: Puerariae Radix juice 50%,sugar 5%,oligoisomaltose 0.5%,β-cyclodextrin 0.15%, puerarin flavonoids extract 0.05%,Vc0.002%.This paper discusses the design of puerarin in soft capsule.
