ObjectiveTo test the expression of NMDAR in the pancreas of fetal rats. By observing the effect of intrauterine hypoxia on the calories of food of pregnant rats, survival rate, birth weight, placenta weight, the ratio of placenta weight to birth weight, pancreas weight, the ratio of pancreas weight to birth weight, the fraction ofβcells in the pancreatic islet of fetal rats, and the effect of MK-801 on the dystrophy of the pancreas of fetal rats induced by intrauterine hypoxia, we try to get to know the function of NMDAR in this process.Methods1. Test the expression of NMDAR in the pancreas of fetal rats by Real-time PCR.2.Observe the effect of MK-801 on the dystrophy of the pancreas of fetal rats induced by intrauterine hypoxia: groups of pregnant rats: the air control group, the air +MK-801 group , the hypoxia group, the hypoxia+MK-801 group. The last three groups are divided into three groups: the 2 days group, the 6 days group, the 10 days group, given treatment on E19,E15,E11, respectively. To get the model of intrauterine hypoxia rats by decreasing the fraction of inspired oxygen of pregnant rats to 10.5±1.0%. Get the fetal rats by opening the uterus on E21. Observing the effect of intrauterine hypoxia on the calories of food of pregnant rats, survival rate, birth weight, placenta weight, the ratio of placenta weight to birth weight, pancreas weight, the ratio of pancreas weight to birth weight, the fraction ofβcells in the pancreatic islet of fetal rats, and the effect of MK-801 on the dystrophy of the development of the pancreas of fetal rats by intrauterine hypoxia.Results1.The effect of intrauterine hypoxia on the development of the entirety and the pancreas of the fetal rats: (1) calories of food of pregnant rats : calories of food of pregnant rats in the hypoxia group is less thanthe air control group(p<0.01 or p<0.05)(2 )the survival rate : the survivalrates of the fetal rats in the hypoxia group are lower than the air controlgroup (p<0.01) (3) birth weight, the rate of low birth weight fetal rats: thebirth weight of the fetal rats in the hypoxia group are significantly lowerthan the air control group(p<0.01),and the rate of low birth weight fetalrats are higher than the air control group( p<0.01). (4) placenta weight,the ratio of placenta weight to birth weight: the placenta weight of thehypoxia group are lower than the air control group (p<0.01), and the ratioof placenta weight to birth weight of the hypoxia group of 6 days arehigher than the hypoxia group of 2 days (p<0.05) , but not different fromthe air control group, the ratio of placenta weight to birth weight of thehypoxia group of 10 days are higher than other hypoxia groups and theair control group (p<0.01) (5) pancreas weight, the ratio of pancreasweight to birth weight: pancreas weight of the hypoxia groups are lowerthan the air control group , and the ratio of pancreas weight to birthweight in the hypoxia group of 10 days (p<0.01) are lower than otherhypoxia groups and the air control groups (p<0.01) (6) the blood sugarof pregnant rats and its fetal rats, and the ratio of the blood sugar ofthem: hypoxia has no effect on the blood sugar of pregnant rats and itsfetal rats. But the blood sugar of the hypoxia group of 2 days are higherthan the control group (p<0.05) ,while the hypoxia group of 6 days and10 days are higher (p<0.01). (7) the morphology of the pancreas and theislets: the number of the islets in the pancreas, the ratio of the pancreasarea to the islet area, number of the islet cells in single islet, the diameterand area of single islet of the hypoxia group of 2 days and 6 days are notdifferent from the air control group. but the ratio of the pancreas area tothe islet area, number of the islet cells in single islet, the diameter andarea of single islet of the hypoxia group of 10 days are less than the aircontrol group (p<0.01 or p<0.05) .HE stain show the dystrophy of thepancreas ,and there is no formation of classic islet-like structure . (8) thefraction of theβcells in the islet: the fraction of theβcells in the islet ofthe hypoxia groups are less than the air control group (p<0.01 orp<0.05). 2. The expression of NMDAR in the pancreas of fetal rats : there are the expression of NMDAR1, NMDAR2A, NMDAR2B, NMDAR2C, NMDAR2D mRNAs, and the expression of NMDAR2D is highest, NMDAR2A and NMDAR2C are less, NMDAR1 and NMDAR2B are lest.3. The effect of MK-801 on the development of the entirety and the pancreas of the fetal rats by intrauterine hypoxia (1) calories of food of pregnant rats: calories of food of pregnant rats in the hypoxia+MK-801 group is the same to the hypoxia group. That of the air+MK-801 group is less than the air control group (p<0.01 or p<0.05) (2) the survival rate: the survival rates of the fetal rats in the hypoxia+MK-801 group are higher than the hypoxia group (p<0.01 or p<0.05) .that of the air +MK-801 group is not different from the control group. (3) birth weight, the rate of low birth weight fetal rats: the birth weight of the fetal rats in the hypoxia+MK-801 group of 2 days are significantly lower than the hypoxia group(p<0.01), and the rate of low birth weight fetal rats are higher than the hypoxia group( p<0.01) .but the birth weight of the fetal rats in hypoxia+MK-801 group of 6 days and 10 days are higher than the hypoxia group(p<0.01),and the rate of low birth weight fetal rats in the hypoxia+MK-801 group of 6 days are lower than the hypoxia group(p<0.05),while there is no difference between the hypoxia+MK-801 group of 10 days and the hypoxia group. The birth weight of fetal rats of the air+MK-801 group are lower than the air control group(p<0.01 ).The rate of low birth weight fetal rats of the air+MK-801 group are higher than the air control group (p<0.01) (4) placenta weight, the ratio of placenta weight to birth weight: the placenta weight of the hypoxia+MK-801 group of 2 days and 6 days are not different from the hypoxia group, and that of 10 days are higher than the hypoxia of 10 days(p<0.01) .the ratio of placenta weight to birth weight of the hypoxia group of 2 days are higher than the hypoxia group of 2 days(p<0.05), but that of 6 days are not different from the hypoxia group of 6 days . the ratio of placenta weight to birth weight of the hypoxia group of 10 days are lower than the hypoxia groups (p<0.01) and not different from the air control group. The placenta weight of the air+MK-801 group are lower than the control group (p<0.01 or p<0.05), and the ratio of placenta weight to birth weight of the air +MK-801 group are same to the control group (5) pancreas weight, the ratio of pancreas weight to birth weight: pancreas weight of the hypoxia+MK-801 groups of 2 days are lower than the hypoxia group of 2 days, that of the hypoxia+MK-801 groups of 6 days and 10 days are higher than the hypoxia group (all p<0.01 or p<0.05 ) .the ratio of pancreas weight to birth weight in the hypoxia+MK-801 group of 2 days and 6 days are not different from the hypoxia groups and the control groups , but that of the hypoxia+MK-801 group of 10 days are higher than the hypoxia group of 10 days (p<0.01) . the pancreas weight of the fetal rats in all of the air+MK-801 group are lower than the air control group, only the ratio of pancreas weight to birth weight in the air+MK-801 group of 6 days are lower than the air control group (p<0.01) (6) the blood sugar of pregnant rats and its fetal rats, and the blood sugar of them: the blood sugar of pregnant rats and its fetal rats in the hypoxia+MK-801 groups are same to the hypoxia groups, the ratio of the blood sugar of the hypoxia+MK-801 group of 2 days are lower than the hypoxia group of 2 days (p<0.05), and that of hypoxia+ MK-801 group of 6 days and 10 days are higher than the hypoxia groups (p<0.01). the blood sugar of pregnant rats and its fetal rats in the air +MK-801 groups are the same to the control groups, the ratio of the blood sugar of the air+MK-801 group of 6 days are higher than the air control group (p<0.05).(7 )the morphology of the pancreas and the islets: the number of the islets in the pancreas, the ratio of the pancreas area to the islet area, number of the islet cells in single islet, the diameter and area of single islet of the hypoxia+MK-801 groups of 2 days and 6 days are not different from the air control group, but the number of islets in the pancreas of the hypoxia +MK-801 group of 10 days are larger than the hypoxia group of 10 days (p<0.05), and the ratio of the islet area to pancreas area increases . HE stain shows a better development of islets in the hypoxia +MK-801 group of 10 days than the hypoxia group of 10 days, the ratio of the pancreas area to the islet area, the diameter and area of single islet of the hypoxia+MK-801 groups of air +MK-801 groups are lower than the air control group (p<0.01 or p<0.05). (8) fraction ofβcells in the islet: the fraction ofβcells in the islet of hypoxia+MK-801 group of 2 days are not different from the hypoxia group of 2 days. But that of hypoxia+MK-801 group of 6 days and 10 days are higher than the hypoxia groups (p<0.01 or p<0.05), that of the air+MK-801 groups are significantly lower than the air control group (p<0.01 or p<0.05) .Conclusion1. We are the first to find the pancreas of the fetal rats express NMDAR, and the expression of NMDR2D is highest, NMDAR2A and NMADR2C less, NMDAR1 and NMDAR2B lest.2. We make a success model of intrauterine hypoxia.3. MK-801 can improve the dystrophy of the entirety and the pancreas of fetal rats which is induced by prolonged chronic hypoxia. The over activation of NMDAR has critical function in this process.4. MK-801 can induce the dystrophy of the entirety and the pancreas of normal fetal rats, which indicates that NMDAR may has important function in the development of the entirety and the pancreas of the fetal rats.
