Objective:Telomeric repeat binding factor 1(TRF1) and human protection of telomeres 1(hPOT1) are two of most important proteins in the protection and modulation of the length of telomeres.Previous studys have indicated that TRF1 was a negative regulator of telomeric length and hPOT1 was a positive regulator of telomeric length.Our object is to study the expression of TRF1 and hPOT1 in nasopharyngeal carcinoma through quantitative reverse transcription polymerase chain reaction (RT-PCR),which will to help to explore the relationship between this two proteins and the occurrance,development,and radiation sensitivity in nasopharyngeal carcinoma.Method:The technique of quantitative reverse transcription polymerase chain reaction(RT-PCR) is applied to detect the TRF1 and hPOT1 mRNA expression in 47 nasopharyngeal carcinoma tissues,which include 39 radiation sensitive nasopharyngeal carcinoma tissues(39/47), radiation resistive nasopharyngeal carcinoma tissues(8/47),and nasopharyngitis tissues(10/47).Compare mRNA expression levels of TRF1 and hPOT1 in different groups and make statistic analysis.Result:1.the mRNA expression of TRF1 in nasopharyngeal carcinoma tissuesThe relative expression of the TRF1 mRNA is 0.0205,which is lower than that in nasopharyngitis tissues(0.0519) and made no statistical significance(P>0.05);The relative expression of the TRF1 mRNA in radiation sensitive nasopharyngeal carcinoma tissues is(0.0139),which is lower than that in radiation resistive nasopharyngeal carcinoma tissues(0.0230) and made statistical significance(P=0.02).The relative expression of the TRF1 mRNA in group T1+T2(0.035) is higher than that in group T3+T4(0.0146 P=0.047);the difference between stageⅠⅡ(0.0342) and stageⅢⅣ(0.0228) has no statistical significance (P>0.05);the difference between group N1+N2(0.0342)and group N0 (0.0205) also has no statistical significance(P>0.05).2.the mRNA expression of hPOT1 in nasopharyngeal carcinoma tissuesThe relative expression of the hPOT1 mRNA in nasopharyngeal carcinoma tissues(0.0014)is higher than that in nasopharyngitis tissues(0.0007 P=0.005);the difference between radiation sensitive nasopharyngeal carcinoma tissues(0.0013)and radiation resistive nasopharyngeal carcinoma tissues(0.0017) made no statistical significance(P>0.05).The relative expression of the hPOT1 mRNA in group T1+T2(0.0012)is lower than that in group T3+T4(0.0016 P=0.045); The relative expression of the hPOT1 mRNA in stageⅠⅡ(0.0011) is lower than that in stageⅢⅣ(0.0014 P=0.022);The relative expression of the hPOT1 mRNA in group group N0(0.0010) is lower than that in group N1+N2(0.0015 P=0.016).3.The mRNA expression of TRF1 and hPOT1 in nasopharyngeal carcinoma has no distinct linear correlation(r=0.019,P>0.05).Conclusion:1.The expression of TRF1 in radiation resistive nasopharyngeal carcinoma is lower than that in radiation sensitive nasopharyngeal carcinoma.TRF1 might be a valuable molecular marker for judging whether there will be radiative resistance.2.hPOT1 expression in nasopharyngeal carcinoma tissues is obviously higher than that in nasopharyngitis tissues and up-regulation of hPOT1 is related to the T and clinic stages and whether there is lymphocyte metastasis,which provde the evidence that it might relates to the occurrance,development of nasopharyngeal carcinoma.
