Objective:The purpose of this study is to examine the expression of XIAP and Caspase-3 in osteosarcoma and trying to discuss the relationship of XIAP and Caspase-3 in osteosarcoma,investigate the possible role of XIAP and Caspase-3 in osteosarcoma and further explore the pathogenesis of osteosarcoma.Method:Specimens of 35 osteosarcoma and 15 Osteochondroma who underwent tumor resections in our hospital and had complete clinical details were fixed in 10%formalin and routinely embadded in paraffin.Then all specimens were examined the expression of XIAP and Caspase-3 by immuno-histochemical method using Monoclonal antibodies of XIAP and Caspase-3,and scored them under high-power microscopy.Put the data into SPSS13.0 and analyze the correlation between the expression level of XIAP and Caspase-3 and the differentitation,metastasis and other clinical features of osteosarcoma.Result:1.In 35 osteosarcoma the expression rate of XIAP was 77.14%(27/35) Comparing with the expression rate 46.70%of Osteochondroma,It is significant higher than that in osteochondroma (P<0.05),and the expression of XIAP in osteosarcoma has no obvious relationship with clinicopathology characteristics(P>0.05).2.The expression rate of Caspase-3 in osteosarcoma was 54.29%,which is significant lower than that in osteochondroma 86.70%(P<0.05),but related to tumor cell differentiation(P<0.05).3.The expression of XIAP was inversely correlated with Caspase-3(r=-0.398,P<0.05).Conclusion:1.The rate of XIAP expression is up-regulated in osteosarcoma and the rate of Caspase-3 expression is down-regulated in osteosarcoma.This study indicates that XIAP and Caspase-3 might be closely related to the carcinogenesis and development of osteosarcoma.2.The expression of Caspase-3 may have relatinship with the degree of malignancy of osteosarcoma.
