Objective To determine the contents of interleukin 8(IL-8) and interleukin 10(IL-10) in the herniated and non-herniated lumbar: intervertebral discs, and to investigate the relationship between IL-8, IL-10, and disc herniation and sciatica.Methods The tissues of the herniated discs were obtained from 29 lumbar disc heraiation(LDH) patients in the Spine Surgery Department of Xiangya Second Hospital from February 2008 to July 2008. Samples were divided into the following three groups: Annulus Intact(AI) group(n=10), Extruded(EXT) group (n=10) and Sequestrated(SEQ) group(n=9), 19 males, 10 females, aged 19-54y. As non-herniated group, 10 normal lumbar discs were harvested from 7 patients(5 males, 2 females, aged 15-41y), 3 lumbar fracture and 4 idiopathic scoliosises. LDH patients' sciatica were evaluated by a visual analog scale (VAS) preoperatively. After harvesting all the samples, make them into tissue homogenates, save the supernatant liquids. Determine the contents of total protein of each sample by ultraviolet absorption method, then Determine the contents of IL-8, IL-10 by enzyme-linked immunosorbent assay(ELISA). Compare the differences between ratios of IL-8, IL-10 and total protein. Relations between IL-8, IL-10 and VAS were carried out by rank correlation analog. Result IL-8 was detected in both herniated and non-herniated group, but IL-10 was only detected in experimental group. Contents of IL-8 in herniated group were significantly higher than those in non-herniated group(P<0.01). Contents of IL-10 in herniated group were higher than those in non-herniated group(P<0.05). Contents of IL-8, IL-10 in herniated group were not increased accompanied with the increase of disc herniation, But contents of IL-8, IL-10 in the EXT and SEQ(annulus not intact) were significantly higher than those in non-herniated and AI(annulus intact)(P<0.01). Rank correlations were found beween IL-8&IL-10, IL-8&VAS, IL-10&VAS (r_s>0, P<0.05) .Conclusion Herniated lumbar intervertebal discs can produce IL-8 and IL-10, contents of IL-8 and IL-10 were significantly increased after the disruption of the annulus fibrosus, accompanied with the increase of sciatic, indicate that IL-8 and IL-10 play a role in LDH and the producing and regulating inflammatory pain.
