The Study About Mechanism Of STAT3 And VEGF In Psoriasis

Objective: To study and research the expression and relationship of signal transducers and activators of transscription (STAT3) and vascular endothelial growth factor (VEGF) in psoriasis vulgaris, and to investigate the mechanism of psoriasis.Method: 20 patients with psoriasis vulgaris were observed. The severity of symptom of psoriasis was assessed by PASI score .The expression of STAT3 and VEGF were detected by Streptavidin-Peroxi dase immunohistochemical method, and the concentration of VEGF in sera was measure by double antibody sandwich enzyme-linked immunosorbent assay (ELISA) in patients; And the 20 normal specimens as control.Result: 1. Serum concentration of VEGF in psoriasis was 790.05±155.21 pg/mL, which significantly higher than in the normal control (487.45±70.66 pg/mL)(P < 0.01).2. VEGF was major expressed in the plasma of basal layer to granular layer of keratinocyte in lesinoal epidermis, while it was almost existed in normal epidermis. In lesional epidermis, STAT3 was expressed in the whole layers or in the cytoplasma and nuclei of most keratinocytes. In normal skin tissues, the stain of STAT3 was lowly located in the cytoplasma. The percentage of positive stain cells of VEGF and STAT3 proteins in the patients'lesions were both higher than in the normal (P < 0.01).3. There were a positive correlation between the level of serum VEGF with the scores of VEGF in the lesion of patient(r=0.872, P < 0.01),and the expression of VEGF was significant correlation with STAT3 in the Lesions ( r=0.822, P < 0.01) . 4. There were a positive correlation between the scores of STAT3 in the lesions with PASI(r=0.498,p<0.05), while not between the level of serum VEGF and VEGF stain score.Conclusion: 1.The level of STAT3 and VEGF in the epidermis of patients with psoriasis vulgaris was abnormal.2.The positive correlation of STAT3 and VEGF suggested that the both might play an cooperation role in psoriasis. 3.STAT3 may indicated the seriousness of the psoriasis lesions indirectly.
