Objective:The purpose of this study was to investigate the expression of the TSLC1 tumor suppressor gene, estrogen receptor (ER), and progesterone receptor (PR) and to understand the expression relationship and clinical significance between TSLC1,ER and PR in breast carcinoma.Method :Immunohistochemistry (SP) was used to detect the expression of ER, PR and TSLC1 in 50 breast carcinomas and nearby no-tumorous tissues.Results:1. The positive expression rate of TSLC1 in breast carcinoma [40% (20/50)] was significantly lower than that in nearby no-tumorous tissues [92% (46/50), P<0.05]. The positive expression rate of TSLC1 in carcinoma in situ and invasive breast carcinomas was 85.7%(6/7)and 32.5% (14/43) respectively (P<0.05), showing a decreasing tendency of positive rate with the advancing invasion. There was also significant difference of TSLC1 protein expression between breast carcinomas with lymph node metastasis [22.7% (5/22)] and without lymph node metastasis [53.5% (15/28), P<0.05]. No significant correlation was found between TSLC1 expression level and patient age or tumor size or tumor stage(P>0.05).2. Positive expression rates of ER [44.0%(22/50)] and PR [42.0% (21/50) ] in breast carcinomas were found no difference(P>0.05).The positive expression rate of ER in carcinoma in situ and invasive breast carcinomas was 85.7%(6/7)and 37.2% (16/43) respectively (P<0.05).The positive expression rate of PR in carcinoma in situ and invasive breast carcinomas was 85.7% (6/7)and 34.9% (15/43) respectively (P< 0.05), showing a decreasing tendency of positive rate with the advancing invasion. There was significant difference of ER expression between breast carcinomas with lymph node metastasis [27.3%(6/22)] and without lymph node metastasis [57.1% (16/28), P<0.05].There was also significant difference of PR expression between breast carcinomas with lymph node metastasis [22.7%(5/22)] and without lymph node metastasis [57.1%(16/28),P<0.05].3. The coexpression of TSLC1 protein and ER was 13, negative expression of both was 21 in 50 breast carcinomas, the common consistency was 68%, and positively correlated(r_s =0.345 ,P<0.05).4.Xhe coexpression of TSLC1 protein and PR was 12,negative expression of both was 21 in 50 breast carcinomas, the common consistency was 66%, and positively correlated (r_s =0.298 ,P<0.05).5.The coexpression of ER and PR was 13 in 50 breast carcinomas, negative expression of both was 20, the common consistency was 66%, and positively correlated(r_s =0.307, P< 0.05). CONCLUSION:1. The positive expression rate of TSLC1 was significantly lower than that in nearby no-tumorous tissues and the positive expression rate had a decreasing tendency with the invasion advances(from carcinoma in situ to invasive carcinoma), indicating that the down expression of TSLC1 is associated with the initiation and development of breast carcinoma.2. The imbalanced expression of ER and PR were observed in breast carcinomas, indicating that the expression of ER and PR is associated with the biological behavior of breast carcinoma. the positive expression rate of ER and PR had a inverse relation with the invasion advances. With the tumor stage development, the positive expression rate ER and PR would be more lower .the positive expression rate of ER and PR had a inverse relation with the lymph node metastasis . With the higher expression of ER and PR, the lymph node metastasis rate would be more lower.3. The positive expression of TSLC1 protein was positively correlated with the expression of ER and PR. These results indicated that the downregulation of TSLC1 and the imbalanced expression of ER and PR in breast carcinomas played an important role in the development and progression of the breast carcinomas.
