ChapterⅠEffect of salvia miltiorhiza on the Movement of Orthodontic Tooth and Expression of bFGF within PeriodontalTissuesObjective To explore the interaction between orthodontic tooth movement and basic fibroblast growth factor(bFGF).To investigate the effect of salvia miltiorhiza on the expression of bFGF within periodontal tissues,thus to propose the possible mechanisms that salvia miltiorhiza accelerate orthodontic tooth movement.Methods 45 rabbits were randomly divided into 3 groups(groups A,B and C).Groups A and B included experimental group of 1,3,7 and 14 days respectively,with 5 rabbits in each group.We observed,the mandibular firstmolar of each experimental rabbit.The rabbits of groups A was designed orthodontic appliance,by which 60g mesial force was applied to pull the mandibular tirst molars;additionally accompanied by injection on buccalgingice of the mandibular first molars with salvia miltiorhiza into the right(grouop A-R) and with normal sodium into the left as the control(group A-L).Groups B was not designed orthodontic appliance,and only received injection with salvia miltiorhiza into the right(grouop B-R) and with normal sodium into the left as the control(group B-L). Group C was not injected and was not designed orthodontic appliance as the control.After killed on schedule,each rabbit of all groups was measured the amount of experimental tooth movement and made into the sections of peridontal tissues.The sections were studied hiostologically by HE stain and examined the expression of bFGF by immunohistochemical method.Results The amount of experimental tooth movement increased successively from 1 to 14 days.The difference among day 3,7and 14 was significant in the comparison between group A-R and group A-L(P<0.01).Histological studying results:Groups B-L and C were normal.Group B-R was only found vessel reaction without bone remodeling.Groups A-R and A-L were found new collagenous fibers,neovascularization,osteoclastic bone resorption on the pressure side and osteoblastic new bone formation on the tension side,which increased successively from 1 to 7 days but decreased comparely on day 14.These changes were significantly lareger in group A-R than group A-L.3,Immunothistochemical result:The expression of bFGF in groups A-R,A-L and B-R enhanced apparently compared with that in groups C and B-L(P<0.01),but that in group A-R is the most apparent(P<0.01),The epression of bFGF reached the peak leve on day 3 in groups A-R and B-R(P<0.01),but it eached the peak level on day 7 in groups A-L and B-L(P<0.01).In all groups,the expression of bFGF was found in the cytoplasm of fibroblast,vascular endothelial cells,osteoblast and cementoblast,but was not found in osteoclast.Conclusions 1,Salvia miltiorhiza may acclelrate orthodontic tooth movement.2,The expression of bFGF is enhanced and bFGF may play an important role in the remodeling of periodontium in the earlier period of orthodontic tooth movement.3,The mechanism that salvia miltiorhiza acclelerate orthodontic tooth movememnt may be salvia miltiorhiza up-regulate the expression of bFGF in periodontium in the earlier period of orthodontic tooth movement.ChapterⅡEffect of basic fibroblast growth factor on proliferation and dose-effect relationship of periodontal ligament cells in vitroObjective To detect the effects of bFGF on proliferation of human periodontal ligament cells in vitro,thus to explor the mechanisms for bFGF to join orthodontic tooth movement.Methods Human periodontal ligament cells were cultured in vitro,and the 5th passage of cells planted in 50 wells of a 96- well culture plate and every well contained 4×10~3 cells.They were divided into 10 groups and every group contained 5 wells.According to bFGF concentration,no bFGF group,0.5ng/ml bFGF group,1.0ng/ml bFGF group,2.0ng/ml bFGF group,5.0ng/ml bFGF group,10.0ng/ml bFGF group,20.0ng/ml bFGF group,30.0ng/ml bFGF group,40.0ng/ml bFGF group and 50.0ng/ml bFGF group were named respectively. When all groups being in exponential phase of growth under microscope,they were dyed by MTT method and OD value of every well was detected by enzyme linked immunodetection meter.Results By contrast to OD mean of control group,OD mean of every group increased gradually from 0.5ng/ml to 20.0ng/ml of bFGF concentration(P<0.05),and reached the peak level on 20.0ng/ml of bFGF concentration.While from 30.0ng/ml to 50.0ng/ml of bFGF concentration,OD mean of every group decreased step by step(P<0.05).Conclusions bFGF may play an important role in rebuilding of the periodontal membranes,bFGF with lower concentration can promote the growth of human periodontal ligament cells;but higher concentration can inhibit the growth of human periodontal ligament cells.
