Part-1 :The Establishment of Transplanted Hepatic CancerModels in SD Rats and its MRI-property Objective: To elucidate the feasibility of the establish an ideal SD-rat model of transplanted hepatic cancer suitable for interventional therapy and magnetic resonance imaging (MRI).Methods: The male weaning SD rats were inoculated and transgenerated from the transplantation tumor rats (Walker-256) by intraperitoneal injection of abdominal fluid of tumor cells, other weaning-SD rats were prepared for the models of solid tumor by subcutaneous inoculation of the abdominal fluid of tumor cells in oxter and groin. After 7 days, these subcutaneous inoculations were successful. The solid tumors were cut to pieces of 0. 2 cm×0. 2 cm×0.2 cm ,which were inserted into the sub capsular pocket of the liver in 17 adult male SD rats , and the transplantation hepatic tumor models were established .After 8 days , MRI was performed for analyzing the tumors growth and its character and the tumors were taken out for pathological examination.Results: The observation of the whole example displayed that the tumor is gray and like fish texture. The dividing line of the tumor tissue and the normal hepatic tissue is distinctness. No central tumor necrosis or hemorrhage was observed by the pathological examination of the sections. The tumor was supplied with plenty of blood vessels. Via pathological examination, the three times of going down to next generation are successful. An isolated and solitary liver tumor was found in 15 rats on MRI,two of them have ascites. The average tumor volume was (0.18±0. 09) cm3 . The tumors were hypointense on T1WI and hyperintense on T2WI.Conclusion: SD rats are economical , the successful rate of model was high ,and optimal effects of MRI for observing the tumor pattern made this tumor model suitable for the study of interventional therapy. Part-2 :Inhibitory effect of angiogenesis inhibitor YH-16 in combination with chemoembolization on rat's transplanted hepatomaObjective: To study the effect of angiogenesis inhibitor YH-16 in combination with chemoembolization on rat's transplanted hepatoma.Methods: The rats bearing metastatic liver cancer of Walker 256 were divided into four groups randomly, with 12 rats in each group。The treatment plan was as follows, group A:normal saline was slowly infused through hepatic artery;group B:supra-liquation iodipin was slowly infused through hepatic artery;group C :mitomycin +supra-liquation iodipin were slowly infused through hepatic artery; group D:mitomycin+ supra-liquation iodipin +YH-16 were slowly infused through hepatic artery. The volume of primary tumor was examined 10 days after drug injection. The expression of vascular endothelial growth factor(VEGF)and tumor microvessel density(MVD)were detected by immunohistochemical method in liver tumors .Results: The volume of tumor in group B, group C and group D was smaller than that in group A (P<0.05), and the volume of tumor in group D was smaller than that in group B and group C(p<0.05),The expression of VEGF and MVD in group B was significantly higher than that of group A(p<0.05). There was no significant difference between Group A and Group C (p>0.05).The expression of VEGF and MVD in group D were significantly lower than that in Group A and group C (P<0.01),showing significance differences .Conclusion:YH-16 infusion through hepatic arteries with transcatheter arterial chemoembolization can suppress the growth of transplanted hepatoma manifestly and it has predominant inhibitive function on the microvessel population of tumor tissue and the expression of VEGF。...
