| 1.Objective:To investigate the relationship between progesterone receptor gene polymorphism,MDR3(multidrug resistance 3) missense mutation and and ICP(intrahepatic cholestasis of pregnancy)2.Methods:Sixty cases of ICP and 60 normal pregnancy women(control group) were investigated,(1)The PROGINS polymorphism of PR gene were detected by polymerase chain reaction method in 60 pregnant women with ICP(ICP group)and 60 normal pregnant women(control group),(2)The missense mutation in exon 14 of MDR3 gene were detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method in 60 pregnant women with ICP(ICP group)and 60 normal pregnant women(control group)。3.Results:3.1 The frequencies of T1/T1,T1/T2,T2/T2 genotypes of PROGINS polymorphism were 90%,10%and 0%respectively in ICP group,and were 91.67 %,8.33%and 0%respectively in control group.There was no significant difference in the distribution of PRGIONS genotypes between the two groups(p=0.7517>0.05);The frequencies of the two alleles T1 and T2were95.0% and5.0%in ICP group,and were95.83%and 4.17%in control group respectively.There was also no significant difference in the frequency of PROGINS alleles between the two groups(p=0.7576>0.05).3.2 The frequencies of GG,GA,AA genotypes in exon 14 of MDR3 gene were 83.33%,16.67%and 0%respectively in ICP group,and were 100%,0%and 0 %respectively in control group,There was significant difference in the distribution in exon 14 of MDR3 gene genotypes between the two groups(p=0.0030<0.05);The frequencies of the two alleles G and A were 91.67%and 8.33%in ICP group,and were100%and 0%in control group respectively,There was also significant difference in the frequency of in exon 14 of MDR3 gene alleles between the two groups(P=0.0036<0.05)。4.Conclusion:4.1 The PROGINS gene polymorphism in the jiangxi Han population may be not associated with the risk of ICP。4.2 The missense mutation of in exon 14 of MDR3 gene may be associated with the the hereditary susceptibility of ICP in jiangxi。... |
