Objective To evaluate the protective effects and mechanisms of ulinastatin in rats with paraquat-induced acute lung injury.Metheds seventy SD rats which have same condition were randomly divided into three groups:paraquat group(PQ group 25), ulinastatin group(UTI group 25) and normal control group(NC group 20).200mg/kg paraquat dissolvent Was poured into stomach of rats bv intragastfie administration in paraquat group and ulinastatin group for once.After twenty minutes,10,0000UI/kg ulinastatin Was injected by peritoneal injection in ulinastatin group,once a day until scarifieated,equal volume normal sodium Was injected by peritoneal injection in the other two groups, once a day until Injection. Were respectively to detect the 8h, 24h, 48h, 72h, 5d five time points in three groups of serums MDA content and SOD,GSH-PX,MPO vitality and the corresponding point in time in BALF cell count, protein content, lung tissue W/D, PaO2 values observed in lung tissue and pathological changes.Resultsâ‘ Serum MDA,MPO,SOD and GSH-PX dynamic changes: PQ group and UTI serum MDA content, MPO vitality were higher than those in NC group (P<0.05). SOD and GSH-PX in serum were All decreased (P<0.01). Compared with NC group,the level of MDA and MPO in PQ group and UTI group increased Significantly both in serum (p <0.05); SOD and GSH-Px in serum were All decreased (P<0.01). The activities of SOD and GSH-PX were all decreased,Compare UTI group with PQ goup,the MDA level decreased both in Plasma and BALF (p<0.05),the SOD and GSH-PX activities were increased as well (p<0.05).â‘¡BALF cell count, protein content, lung tissue W/D: PQ group and the UTI group BALF cell count, protein content, lung W/D higher than the NC group; UTI group than in the PQ group, the differences were significant (P<0.01).â‘¢PaO2: Compared with NC group,PQ group and the UTI group PaO2 lower , and the UTI group than the PQ group PaO2 increase was significantly higher (P <0.05).â‘£Change of pathological section: there was no alteration in lung tissue in NC group. To observe the whole lung was pink, no hyperemia, edema and infarction. In PQ group severe congestion and lung edema and the lung swelled. There was appear dark red lung, lung volume increased significantly hyperemia, edema, showing a wide range of focal points, or bleeding, visible incision pink bubble like liquid overflow. In UTI group there were slight alteration compared with that in injury group. Light microscope(HE): PQ group shows that focal pulmonary hemorrhage, alveolar wall thickening apparent, alveolar and interstitial pulmonary edema, inflammatory cell infiltration, and some sections can be seen focal atelectasis, emphysema. UTI Group hemorrhage, edema, inflammatory cell in filtrat- ion than the change in PQ group.Conclusion: 1. PQ rats obvious in the state of oxidative stress, over-reaction of lipid peroxidation involved in the occurrence of acute lung injury in rats.2.UTI reduce paraquat poisoning caused by diffuse lung injury, inhibiting lipid peroxidation, reducing oxygen free radical damage, PQ significantly improve the state of oxidative stress in rats, thus PQ-induced lung injury has a protective effect .
